Capillary electrophoresis (CE) is one of the most active domains in the separation science. However, the poor concentration sensitivity of CE is a crucial point for trace analysis occurring in very low concentration. A number of on-line concentration techniques have been developed to increase the sensitivity of CE. On-line concentration techniques have been employed more and more in many analytical areas over the past decade due to their advantages of simplicity, automation, low cost and high sensitivity. Therefore, study on the on-line concentration techniques is significant to both the development of CE technique and the extension of its application domain.In this thesis, the CE methods for the separation and determination of the effective components of Sophora flavescens Ait, Panax notoginseng and Fufang Danshen Pian were developed. And, on-line concentration techniques were used to increase the sensitivity of the analytical methods. The main contents are described as follows:(1) A non-aqueous capillary electrophoresis procedure with field-amplified sample stacking (PASS) concentration for the separation and determination of four minor alkaloids in Sophora flavescens Ait was established. By applying FASS for on-line sample concentration, the concentration sensitivity was about 1 order magnitude higher than that obtained by the hydrodynamic injection. The limits of detection were 0.20-0.57μg/mL.(2) A novel micellar electrokinetic capillary chromatography method was developed for the simultaneous separation of 10 saponins in Panax notoginseng. By using field-enhanced sample injection with reverse migrating micelles on-line concentration technique, the limits of detection on standards were 1.9-6.3μg/mL under the optimum stacking conditions. The concentration sensitivity of CE could be enhanced up several decuples compared to the literature method.(3) A reversed migration micellar electrokinetic chromatography method was developed for the simultaneous separation and determination of five saponins in Fuang Danshen Pian. The proposed method was simple, reproducible and promising alternative to analyze and process the quality control of other Chinese medicinal preparations containing saponins.The studies in this thesis demonstrate that the on-line concentration technique is very important for the application and extension of CE to trace analysis due to its great advantages such as simple operation, low cost and high sensitivity.
