The Expression And Significance Of The Matrix Metalloproteinase Inhibitor RECK Gene In Endometrial Carcinoma

IntroductionThe endometrial cancer is one of the most familiar malignant tumors in women.The same as other malignant tumors,its metastases and invasion are the important characteristics and are also the main reasons of the death of patients with carcinoma.During the course of the tumors metastases and invasion,the matrix metalloproteinase(MMPs)is essential.The matrix metalloproteinase(MMPs)pay the most important role in the metastases and invasion.RECK(reversion inducing cysteine rich protein with kazal motifs)is a new matrix metalloproteinase inhibitor,It had been proved that the RECK protein regulates at least three members of the matrix metalloproteinase(MMP)family,MMP-2,MMP-9,and MT1-MMP,to depress tumors metastases and invasion.Study suggests RECK is correlations with the prognosis of patients with carcinoma.The prognosis in those patients who had high expression levels of Reck in tumorous tissue showed significantly better than those had lower expression.So it is very significant to search safe and effective ways to depress the metastases and invasion.In the present study,we used immunobistochemical method to detect expression of RECK in tumorous tissue of endometrial carcinoma,analyzed the relation among the tumorous tissue development,TMN stage and the type of histology.We wanted to afford a new idea for the clinic diagnose,therapy of endometrial cancer and judging the situation of patient after surgical operation. Materials and methods1.Sample preparingThe 43 cases were obtained from The Second Affiliated Hospital Medical University from Jan,2003 to Dec,2006,Which were paraffin-embedded sample of aboard hysterectomy.The type of histology are all endometrial adenocarcinoma;The pathological grade G₁ 11 cases,G₂ 18 cases,G₃14 cases;Superfical musle invasion(＜1/2=30cases,deep musle invasion(≥1/2)13cases;lymph transfer 12 cases,without lymph transfer 21 cases;operation pathology stages(FIGO,2000):â… 17 cases,â…¡11 cases,â…¢15 cases, 20 cases paraffin-embedded sample of normal endometria at the same period were comparision group.All the cases didn't accept radio-or chemical-therapy before surgery.2.Mainly agentRECK anti-human monoclonal antibody of the rat,SP staining kit and DAB kit were both bought from MBL Company.The chroma of first antibody is 1:1003.Empirical method and resultAssessment using SP immunohistochemical method to detect the expression information of RECK in endometrial cancer.Immunohistochemical positive is defined as yellow drops in cell membranes.Count the positive cells less than 10%,positive cells 10%～24%,positive cells 25%～49%,positive cells more than 50%,Which were parting graded:0,1,2,3chon,The dying intention was set off 4 grades:achromatism,buffy, saffron,brown were parting graded:0,1,2,3.Two grades were added together.0～3was negative,more than 3 chon was positive.4.Statistical analysisUse SPSS11.5software,All data uses X² test.α=0.05 was considered to be significance. Result1.RECK positive staining mainly locates in cell membrane.weak-positive expresses in tissue of endometrial carcinoma,expression rate is 51.2%.However, expression is obviously enhanced in normal endometria,expression rate is 80.0%. There is significant deviation between two groups(P＜0.05).2.The positive expression rate of RECK inâ… stage cancer tissue is obviously higher thanâ…¢stage of endometrial carcinoma,There is significant deviation between two groups(P＜0.05).however there is not significant deviation betweenâ…¡stage andâ… stage(P＞0.05).There is significant deviation betweenâ…¡stage andâ…¢stage(P＜0.05) The positive expression rate of RECK in superfical musle invasion is obviously higher than deep musle invasion of endometrial carcinoma.There is significant deviation between two groups(P＜0.05).The positive expression rate of RECK is not significant deviation among the pathological grades of endometrial carcinoma.also there is significant deviation between the two groups of lymph transfer and without lymph transfer..RECK(P＜0.05).Conclusion1.The expression of Reck had negative correlation with the operation pathological stages of endometrial carcinoma2.RECK express abroadly in normal endometria.However,expression is obviously depression in tissue of endometrial carcinoma.3.RECK may inhibit the progress invasion and transfer of endometrial carcinoma.
