With the development of society, the white-collars living in the big cities become the High-risk population of depressive illness under the circumstance of high pressure and high competition. Depression is a group of mood disorders or affective disorder caused by a variety of reasons with main symptoms by depressing and is a group of clinical symptoms or condition centered by depressive mood and self- experience. In the West, depression was known as the "blue worry". According to the survey, the incidence of depression in developed countries was rose to 8 to 10 per cent in 2005. Based on the latest survey of the World Health Organization, the incidence of depression is about 3.1% in globe, while which is close to 6 percent in developed countries. In 2002, the patients with severe depression have more than 8,900 million people and the depression patients have reached 340 million in globe. In adult population older than 20-year-old, the patients with depression is increasing at a rate of 11.3 percent each year.Meanwhile, with the improvement of living standards, the diabetes complication has become the third group of diseases after cancer and cardio-cerebral vascular diseases that affects people's physical and mental health seriously. The complication of diabetes is the most important factor determining the prognosis of patients and is the most important cause that increases the rate of diabetes mortality and disability. Therefore, the prevention of all kind complications of diabetes has become an important issue in research of diabetes in medicine institutes of the world at present. For the benefit of research and prevention of the complications of diabetes, it is necessary to classify them. But, it is quite difficult to reasonable classifying because that the kinds of complications of diabetes are in wide scope with diverse manifestations in clinical and the etiology and pathogenesis is complex and has not yet been fully clarified. The advanced glycation end products (AGEs) are a variety structure of non-reversible polymers formed after a series of non-enzymatic reaction of protein-free and reducing sugar (such as glucose). And it is thought in a number of studies that the formation of AGEs plays a very important role in the occurrence and development of many diseases caused by aging and diabetes such as diabetic nephropathy, vascular disease, cataract by diabetes, arteriosclerosis and Alzheimer's disease.In traditional Chinese medicine theory, Morindae officinalis can invigorate the kidney, strengthen the bones and muscles and eliminate rheumatism; Picrorhizae has the effect of clearing asthenic fever, cooling blood to stop bleeding, heat-clearing and deprive the evil wetness, heat-clearing and detoxicating, anthelminthic and vanishing malntrition. At the same time, the modern and clinical medical experience proves that Morindae officinalis it has the effects of anti-depression, immunity strengthening, promoting the development of ossature, anti-inflammatory and anti-aging, etc; Picroside has the effects of liver protecting and cholagogue, antibacterial and anti-inflammatory and anti-ischemic.In this paper, the quality standards of Morindae officinalis were established by experiment and analysis the assaying result by finger printing similar calculation. The active constituents of Picroside are sieved and separated:1) Some Morindae officinalis from different metropolis are researched based on their actual curative effect and the quality specification of Morindae officinalis is established from the aspects of trait, identification and assaying.By the fingerprint similarity calculation software written in MATLAB, the fingerprint of oligosaccharides was established in the first time. Simultaneously, the assaying results of quality standards were analyzed by the fingerprint data processing methods to explore the quality changes rule of the root bark of medicinal Morinda officinalis in the process of growth.Furthermore, we adopt the HPLC-MS method to consummate the technical standard system and explore to establish the quality control model of combining the source of Chinese herbal medicines, the production process and detection indicators, which are used to providing the reliable and scientific theoretical guidance in standardized production of Chinese herbal medicines.2) The active sites of Picroside were sieved and its chemical constituents were separated.Based on the producing mechanism of AGEs, the Picroside, the medicine with AGEs activity inhibiting, was preliminary sieved from 12 traditional Chinese medicines with activating blood circulation to dissipate blood stasis and antioxidant. Then, the chemical constituents were separated by using some research methods of chemical plants, such as thermal extraction, liquid-liquid distribution, positive and negative phase silica-gel adsorption and gel-layer chromatography. Finally, the structure of picroside II was identified using the spectral analysis methods of MS, UV, NMR. Finally, the work done in this paper was summarized, which provides some reference for the follow-up study of Morinda officinalis and Picroside.
