Expression Of CD127 In Peripheral Blood Of Patients With AIDS

Objective To investigate the changes of CD4,CD8,CD3,NK,B,CD8~+CD127~+CD3~+cells in peripheral blood of patients with AIDS,and to compare it with healthy controls.And find the reasons why HIV infected individuals have immunodeficiency.Methods chose 194 people(123 AIDS patients and 96 HIV infected individuals without syndromes)who had confirmed by Western Blot.We observed the immune conditions on 33 AIDS patients during the treated period.Then we measured the expression of CD4,CD8,CD3,NK,B,CD127 ~+CD8~+CD3~+ cells in those 194 patients and 30 healthy adults.We chose 8 patients and observed the change of CD127~+CD8~+CD3~+ cells during the HAART treatment.All of the numerical data is handled with SPSS13.0 statistics software.Results The percentage of CD4~+ cells in patients with AIDS was lower than that of healthy adults(p＜0.01);The percentage of CD8~+ cells was higher than that of healthy adults(p＜0.01),CD4~+/CD8~+ was lower than that of healthy adults(p＜0.01),The percentage of CD3~+ cells is higher than that of healthy adults(p＜0.05),NK cells was approached that of healthy adults(p＞0.05),B cells was lower than that of healthy adults(p＜0.05),CD127~+CD8~+CD3~+ cells was lower than that of healthy adults(p＜0.01),the GM of CD127~+CD8~+CD3~+ cells also lower than that of healthy adults(p＜0.01).The percentage of CD127 ~+CD8~+CD3~+ cells in patients treated with HAART was higher than the untreated (p＜0.01).After three months treated with HAART on 33 AIDS patients,we collected the results and handled it with paired-samples t test.CD4,CD8,CD3 cells count in patients treated with HAART was significant increased than those in untreated(P=0.000),but the changes on NK cells,CD4~+/CD8~+ were not significantly(P=0.710),the percentage of B cells after therapy was lower than that of before(P=0.025),the percentage of CD127~+CDS~+CD3~+ cells after therapy was higher than that of before(P=0.001).The pearson correlation of CD127~+CDS~+CD3~+ cells and CD4 count was 0.281,p＜0.05,have significant correlation,but have no significant correlation between CD127~+CD8~+CD3~+ cells and CD8 count or CD3 count.The pearson correlation of CD127~+CD8~+ CD3~+ cells and CD4/CD8 was 0.318,p＜0.05,have significant correlation. Altered CD127~+CD8~+CD3~+expression with aging,p=0.004,have significant different.Conclusions Patients with AIDS have lower immune activity than that of healthy people.After a HAART therapy,the immune activity had reversed but still lower than healthy people,HAART is a effective one in AIDS therapy, decreased CD127~+ expression on CDS~+CD3~+ cells from HIV-infected patients correlates with disease progression and immune activation,the level of CD127 ~+CD8~+CD3~+ cells was correlated with CD4 T cell count,the decreased of CD127 on CD8~+CD3~+ T cells might be a reason for immunodeficiency in AIDS. There is altered CD127~+CD8~+CD3~+ expression with aging.
