Expression Of VEGF,CD44V6 And CD62P Protein In Nasopharyngeal Carcinoma And Its Clinical Application

Objective To investigate the expression of VEGF,CD44V6 and CD62P in nasopharyngeal carcinoma,as well as the relationship and prognostic significance of them.Some new molecularly targted agents are available in the treatment of nasopharyngeal carcinoma.Methods Serum specimens were collected before treatment from 60 nasopharyneal Carcinoma(NPC)patients and 40 healthy control subjects.26 cases with nasopharyneal carcinoma and samples of nasopharyngitis tissues were included in this work.Serum and biopsies VEGF were measured via the quantitative enzyme-linked immunosorbent assay(ELISA)method.Serum and biopsies CD44V6,CD62P were measured via flow cytometry(FCM)method.Through follow-up study,the relationship between VEGF,CD44V6,CD62P and prognosis of NPC patients was investigated.Results 1)Serum and biopsies VEGF,CD44V6,CD62P level of NPC patients is significantly higher than that of control group(P＜0.001).2)Expression of serum VEGF in NPC with or without lymphnode metastasis and distant metastasis was significantly different(P＜0.001),There is no significant dependency between patients' clinical stage(P＞0.05).3)Expression of peripheral blood CD44V6 and CD62P of NPC patients were highly correlated with clinical stage,lymphnode metastasis and distant metastasis(P＜0.05).4)In NPC inⅢ-Ⅳtumor grade,with or without lymphnode metastasis and distant metastasis,expression of tumor biopsies VEGF,CD44V6 and CD62P was increased,but there is no significant dependency between them(P＞0.05).There is no significant dependency between CD44V6 and CD62P level in NPC patients'gender and age(P＞0.05).5)Other clinicopathological features are significance with VEGF,CD44V6 and CD62P(P＜0.05).6)The 1-year,3-year and 5-year recurrence,metastasis and survival according to the expression of VEGF,CD44V6 and CD62P had statistical difference(P＜0.01);With over expression of the 3 factors than in these patients with over expression of lor 2 factors or with low expression of the 3(P＜0.01).Conclusions 1)The VEGF,CD44V6 and CD62P have a positive correlation with the invasion and metastasis of NPC possibly.2)There is a positive correlation between VEGF,CD44V6 and CD62P.3)CD44V6 and CD62P may be helpful to predict the recurrence and metastasis of NPC patients.The assessment of VEGF level in combination with CD44V6 and CD62P level could stratify patients into groups with different potentials of NPC recurrence and metastasis more accurated than CD44V6 and CD62P individually.4)Since CD44V6 and CD62P played pivotal roles in tumor angioensis,development and occurrence of NPC.They would be valuable prognostic markers and serve as potential targets for therapy.
