The Experiment Research Of HTK Solution And UW Solution On Biochemical Function Of Rat Small Intestine Preservation

Objective: To study the effect and disparity of the preservation of rat small intestine with HTK solution and UW solution.Methods: According to preservation solution, the SD rats were divided into three groups randomly: UW group,HTK group and NS group. One group haves twelve rats. Before the experiment, the rats were in abrosia twelve hours, but can have water. All the rats were in narcotism by injecting 3% pentobarbital sodium into cavum abdominis(30mg/kg). The intestine was perfused by intubation via abdominal artery. One rat was perfused preservation solution about 50ml-60ml. when the color of intestine charged to white ,the jejunum of each rat was harvested into two segments , one segment was 6cm, the gut was flushed with metronidazole solution(℃,0.5%) ,and then the intestine segments were preserved in HTK solution,UW solution or NS solution. Determination of lactic acid at 0h,4h,8h,16h and energy materia at 8h,16h were used to evaluate the result.Results: Compared with UW group, lactate contents of HTK group were significantly lower at 4h and higher at 16h (P<0.05). ATP contents of HTK group were significantly higher than UW group at 8h (P<0.05). All variables of both groups were better than the rat intestines by the normal saline group.Conclusion: HTK solution was effective in preservation of the rat intestine. Before 8h, the preservation effect of HTK solution was better than UW solution. But after 8h, the effect of HTK solution was worse than UW solution.
