Radix Astragali is one commonly used traditional Chinese medicine,usually being known as "the best of all medicine of tonifying Qi".It has decisive position in traditional Chinese medicine and has many curative effects.In recent years,with the increase of Radix Astragali consumption,the traditional methods of Radix Astragali usage and processing and active components extraction have been unable to meet the needs of the community.In combination with the modern bio-engineering fermentation technology on the basis of the zymotechnics of traditional Chinese medicine processing,we utilized Pleurotus ostreatus to ferment Radix Astragali.The objection of our work is to achieve its high efficacy and the actual utilization.The results showed that:Using the single-factor of plate growth experiment and liquid flask shaking experiment,the optimal composition of the cultrue medium for Radix Astragali is:Radix Astragali(60 eyes)50g, bran(cook juice)20g,gypsum(60 eyes)1g,distilled water 1000ml,pH value was natural.The major active components were detected in the process(the first,third,fifth,seventh, ninth,eleventh,thirteenth,fifteenth day)of Radix Astragali fermentation.Compared with the decoction of Radix Astragali,the contents of soluble polysaccharids and free amino acids were decreased,total flavonoids,saponins and AstragalosideⅣwere increased in the Radix Astragali fermentation supernatant,yet it did not reach a significant level.And the fermentation duration for Radix Astragali should be seven days.The results of pharmacological experiments on the Radix Astragali fermentation supernatant revealed that the high,median dosages of Radix Astragali fermentation supernatant had obvious anoxic resistance activity,anticoagulation,diuresis activity in mice.Further, combined with pentobarbital sodium the high dosage could prolong sleep latency time and shorten sleep sustained time in mice.But when compared with the decoction of Radix Astragali, there was significant difference only on anticoagulation activity(P<0.05).
