Pleurotus ostreatus is known as Oyster Mushroom. It has both edible and medicinal value. In this research, analysed the volatile components from the dried fruiting bodies of Pleurotus ostreatus with GC-MS. There are 20 kinds of substances(Prob > 50%) were detected, mainly including acids(12.184%), aldehydes(9.113%), furans(7.108%), and esters(6.993%) as well as small amounts of ketones(2.226%) and aromatic hydrocarbon(1.693%), etc. In all substances, the Hexadecanoic acid content is the highest, as high as 4.665%. The bioactive metabolites were separated and purified by methods such as silica gel column chromatography, sephadex column chromatography, high-performance liquid chromatography(HPLC), and identified by spectroscopic analyses. Eight compounds were isolated and their structures were identified as(22E,24R)-ergosta-5,7,22-trien-3β-ol(1), 24R-5α,8α-epidioxyergosta-6,22-dien-3β-ol(2), 22 E,24R-5α,6α-epoxyergosta-8(14),22-diene-3β,7α-diol(3), nicotinic acid(4), adenosine(5), mannitol(6), uridine(7), nicotinamide(8). Compounds(3) and(5) were separated from the fruiting bodies of Pleurotus ostreatus for the first time.Measured the content of nicotinamide and ergosterol in products of solid fermentation, liquid fermentation and different growth periods of Pleurotus ostreatus through HPLC. The content of nicotinamide in coral-like phase, mature stage, liquid fermentatin mycelium, bud stage, solid fermentation products, fermentation liquor is 0.071%, 0.033%, 0.018%, 0.014%, 0.006%, 0.004%. The content of ergosterol in mature stage, bud stage, coral-like phase, liquid fermentatin mycelium, solid fermentation products, fermentation liquor is 0.053%, 0.048%, 0.019%, 0.005%, 0.003%, 0.002%.Taking ergosterol as standard sample, total steriods was measured by the method of vanillin acetic acid, using visible light spectrophotometer. The content of total steriods in fermentation liquor, liquid fermentatin mycelium, solid fermentation products, coral-like phase, bud stage, mature stage is 6.51%, 2.06%, 5.53%, 4.56%, 6.25%, 5.92%.
