Pleurotus ostreatus is a common medicinal and edible fungus.The fruiting bodies of P.ostreatus contain various chemical composition such as polysaccharides,steroids,volatile oils and polyphenols,which have many pharmacological effects,such as anti-tumor,anti-inflammatory,anti-virus,immune regulation,anti-oxidation,hypoglycemia,hypolipidemia,liver protection,insecticide,bacteriostasis and so on.Additionally,there are certain curative effects noted in the treatment of femoral head necrosis and Alzheimer's disease.In this paper,the chemical composition,pharmacological activity of P.ostreatus where summarized,in order to provide reference basis for better research and development of P.ostreatus resources,and its application of medicine,health products,food and other related fields.1.Chemical constituents of fruiting bodies of P.ostreatus.By means of silica gel column,ODS column,Sephadex LH-20 column and preparative high performance liquid chromatography,19 compounds were isolated from different polar parts of P.ostreatus.16 compounds were identified by 1H-NMR,13C-NMR,DEPT,MS and other spectral data,including ergosterol(1)?ergost-7,9(11),22-trien-3?,5a,6a-triol(2)?dibutyl phthalate(3)?bis(2-ethylhexyl)benzene-1,2-dicarboxylate(4)?Dipentyl phthalate(5)?diisobutyl phthalate(6)?bis(2-methylheptyl)phthalate(7)?phenylalanine(8)?pyroglutamic acid methyl ester(9)?Formamide(10)?leptosphaerin(11)?1,2-dihydroxymintlactone(12)?a-dimorphecolic acid(13)?4-hydroxy-17R-methylincisterol(14)?volemolide(15)?(1'R,2'R,3'S,4'R)-1,2,4-triazole nucleoside(16).Five compounds were identified in petroleum ether fraction,namely compounds 1,6,13-15.Six compounds were obtained in ethyl acetate fraction,namely compounds 2-5,7 and 12.Five compounds were isolated in n-butanol fraction,namely compounds 8-11 and 16.Compounds 2-15 were isolated from Pleurotus ostreatus for the first time.2.Study on The Proliferation of Four Cells by Different Polar Parts.CCK-8 method was used to study the inhibitory or proliferative effects of ethanol extract,petroleum ether extract,ethyl acetate extract and n-butanol extract of Pleurotus ostreatus on human hepatocellular carcinoma cells(HepG-2),adenocarcinoma human alveolar basal epithelial cells(A549),rat myoblasts(L6)and mouse osteoblasts(MC3T3).The results showed that:The ethanol extract had the best inhibitory activity on HepG-2 tumor cells,IC50 was 35.3?g mL"1,and the inhibitory rate could reach 66.6%at 500.0?g mL-1.The IC50 value of ethyl acetate extract for HepG-2 tumor cells was 42.1?g mL"1,the IC50 value of n-butanol extract on HepG-2 tumor cells was 48.9?g mL-1,while that of petroleum ether extract was weak,with the IC50 value of 88.5 ?g·mL-1,but the inhibition rate could reach 63.7%at 500.0 ?g·mL-1.The extract from different parts of Pleurotus ostreatus had no effect on A549 tumor cells,while the ethanol extract of Pleurotus ostreatus had a slightly better effect on L6 myoblasts proliferation than the positive drug,but the effect of other parts of Pleurotus ostreatus extract was significantly weaker than that of ethanol extract.Ethyl acetate extract of Pleurotus ostreatus had the best effect on promoting proliferation of MC3T3 osteoblasts and was obviously superior to the positive drug.At 250.0?g·mL-1,the cell viability could reach 199.7%,while the extract of other parts had a weak effect.In this paper,the systematic separation of the chemical constituents of Pleurotus ostreatus and the preliminary screening of its pharmacological activities were carried out,which could provide a theoretical basis for the further study of the pharmacological activities of monomer compounds.
