Pleurtus Spodotecus and Pleurotus ostreatus are all fungus for medicinal purpose. Thisresearch applied biological fermentation engineering method to obtain mixed crudepolysaccharide including polysaccharide of pleurotus spodoleucas'hypha and ginseng dregswhich were all called polysaccharide of ginseng and mushroom by using ginseng extractresidue as the main culture medium of Pleurtus Spodotecus and Pleurotus ostreatus. Study onoptimizing the fermentation culture medium, extraction process, content determination andmedicina1activities of the polysaccharide of ginseng and mushroom was carried out. It wasexpected to provide theoretical foundation for exploitation of polysaccharide of ginseng andmushroom.Firstly, we investigated the feasibility of the co-fermentation of Pleurtus Spodotecus andPleurotus ostreatus. Content of crude polysaccharide of ginseeng dand mushroom wasdetermined by phenol-sulfuric acid-method.Second optimize the fermentation culturemedium.Last, extraction process of polysaccharide of ginseng and mushroom was optimized,which involved the dosage of water,extracted time, extracted times as well as alcoholconcentration for precipitation, then L9(34)was selected to design the orthogonal test and thecontent of polysaccharide was considered as the index to filtrate the best extracting method.Pharmacological experiments including acute toxicity, hypoglycemic effect, preventinghyperlipidemia miceU and anntioxRidant aectivitgy werie scarrited eout orn the crude polysaccharide ofginseng and mushroom(the mixed crude polysaccharide of Pleurotus spodoleucas'hypha andginseng dregs, the mixed crude polysaccharide of Pleurotus ostreatus'hypha and ginsengdregs)and crude hypha polysaccharide(the crude polysaccharide of Pleurotus spodoleucas'hypha,the crude polysaccharide of Pleurotus ostreatus' hypha,). Results showed that they allhad no toxicity, and the MTD of them was determined. The above four crude polysaccharidescould not reduce the blood glucose level of mice significantly. Whereas, it was found that theyhad the effect on regulating the level of serum lipids and also could help strengthen theactivities of SOD, decrease the content of MDA of the organs (Liver, kidney, brain) ofhyperlipidemia rats, act the antioxidant function.On the basis of the above results, the polysaccharides of ginseng and mushroom have the efficacy of preventing hyperlipidemia and antioxidant function on the organs (liver, kidney,brain) of hyperlipidemia rats. This study could provide a scientific basis for the developmentof new drugs.
