Objective: To investigate the protein expression of maspin ,bax and p53 in condyloma acuminatum (CA) lesions, to explore their possible roles in pathogenesis and to find the relation between these kinds of protein and the recurrence of CA.Methods: 35 cases of CA specimens were all collected from dermatology department of Qilu hospital of Shandong university. In 21 cases of recurrent CA,14 non-recurrent CA and 10 normal prepuces lesions, protein expression of maspin ,bax and p53 were detected respectively by immunohistochemistry technique. The results were determined by semi-quantities test, and the statistical analysis was managed by SPSS11.5 for windows software.Results:1. The staining of maspin was located mainly in the cytoplasm and nucleus.In normal controls positive cells scattered throughout the whole epidermis with deep color, while in CA lesions scattered in stratum basale and stratum spinosum with light color. The positive rate and expression intensity of maspin protein increased successively from recurrent CA to normal controls (4.76%, 35.71 %and 100%) (P < 0.05).2. The staining of bax was located mainly in the cytoplasm. The positive cells were observed mainly in stratum spinosum with Hghtyellow or brownyellow. The expression rate of bax showed successively 28.57%,64.29% and 70.00% in recurrent CA, non-recurrent CA and normal controls (P > 0.05). The expression intensity of bax in CA was stronger than that in normal controls (P < 0.05), but there was no significant statistic difference in the expression intensity between recurrent CA group and non-recurrent CA group(P > 0.05). 3. The staining of p53 was located mainly in the nucleus. Normal controls were all negative. In CA lesions, positive cells mostly scattered in stratum basale and stratum spinosum. The positive rate and expression intensity of p53 protein reduced successively from recurrent CA to normal prepuces(P < 0.05).Conclusions:1. Down-regulated expression of maspin may play an important role in the pathogenesis and recurrence of CA. According to more down-regulated expression in recurrent CA than that in nor-recurrent CA,We confirmed that down-regulated expression of maspin may contribute to the recurrence of CA.2. Bax may play an important mediating role in the course of cell apoptosis. However, there was no significant statistic difference in the expression between recurrent CA and non-recurrent CA. As a result, we speculate that there is no significant correlation between the bax expression and the recurrence prognosis of CA.3. The over-expressing p53 protein may accelerate cells proliferation and affect the biologic behavior of CA, which may be another reason for the rapid growth and the recurrence of CA. Therefor, detecting p53 protein can be useful as a guide for evaluating the recurrence prognosis of CA.
