Studies On The Chemical Constituents Of Tibetan Medicine Flos Gentianae Stramienae And Criterion Of Gentiana Straminea Maxim In Chinese Pharmacopoeia

Radix Gentianae Stramienae,one of the well-known traditional Chinese drug "Qinjiao", is the dried root of Gentiana straminea Maxim.,which belongs to the Faro.Gentianaceae.It can relieve rheumatic conditions,and remove damp-heat.Flos Gentianae Stramienae,the dried flower of Gentiana straminea Maxim,is considered as one of the high grade Tibetan drugs with outstanding effectiveness of clearing away heat and toxic materials.In general,its traditional clinical usage is to cure inflammation of stomach and intestines,hepatitis, cholecystitis,etc.Several isolation methods were used for a preliminary research on chemical components of Flos Gentianae Stramineae produced in Qinghai.Nine compounds were achieved at last and they were elucidated by the means of spectroscopy(UV,IR,MS,¹³C-NMR,¹H-NMR,H-H COSY etc.) assisted by comparing with standard substance.They were:isoorientin(â… ), 6'-O-β-D-glucopyranosyl gentiopicroside(â…¡),loganic acid(â…¢),isoorientin-4'-O-glucoside (â…£),β-sitosterol(â…¤),daucosterol(â…¥),gentiopicroside(â…¦),isovitexin(â…§),vitexin (â…¨).Among the totle,vitexin(â…¨) and isoorientin-4'-O-glucoside(â…£) were isolated from genus Gentiana for the first time.On the basis of having systematically isolated chemical composition of Flos gentianae stramineae,five iridoid glycosides and three flavonoids were determined by HPLC and LC-MS,which made an elementary research for further component analysis about Tibetan medicine.The eight component were loganic acid,6'-O-β-D-glucopyranosyl gentiopicroside, swertiamarin,gentiopicroside,sweroside,isoorientin-4'-O-glucoside,isoorientin,vitexin.Meanwhile,we assumed one standard research project of Pharmacopoeia of People's Republic of China Edition 2010:Quality specification research of Radix Gentianae Stramienae.In the experiment,an HPLC method was adopted for the determination of loganic acid and gentiopicroside in medical material and decoction pieces of Radix Gentianae Stramienae of different batches and the content limitation was established simultaneously. Then the contents of loganic acid and gentiopicroside in about thirty-six samples were determined respectively.Further more,we determined the moisture content,ash content and acid-insoluble ash and we optimized the distinguish condition of thin layer chromato^aphy and high performance liquid chromatography.On the basis of the experiment above,the draft of the quality specification of Radix Gentianae Stramienae in 2010 edition Chinese Pharmacopoeia was established.We suggest Radix Gentianae Stramienae new added in future 2010 edition Chinese Phannacopeia.All these work was verified by the Institutions for Drug Control of Qinghai Provience.
