Gentiana straminea Maxim., which belongs to the Gen. Gentiana, Fam. Gentianaceae, mainly distributes at the high altitude of 2000-4950m in Province of Qinghai, Tibet, Gansu and Sichuan, etc. The dried root of G. straminea Maxim., one of the well-known traditional Chinese drug "Qinjiao", is embodied as one of the original plants of Radix Gentianae Stramienae in the China Pharmacopeia (2010 edition), which can relieve rheumatic conditions, and remove damp-heat.Several isolation methods were used for a preliminary research on chemical components of the root of G. straminea Maxim. produced in province Qinghai. Five compounds were achieved finally and they were elucidated by the means of spectroscopy (UV, IR,13C-NMR and 'H-NMR etc.) assisted by comparing with standard substance. They were:β-sitosterol(Ⅰ), roburic acid(Ⅱ), daucosterol(Ⅲ), oleanolic acid(Ⅳ) and gentiopicroside(Ⅴ). Among the totle, roburic acid(Ⅱ) was isolated from G. straminea Maxim. for the first time.On the basis of having systematically isolated chemical composition of the root of G. straminea Maxim., four liposoluble components (oleanolic acid, daucosterol, roburic acid andβ-sitosterol) were simultaneously determined by HPLC-ELSD method, which made an elementary research for further component analysis of comparing G. straminea Maxim. with other plants of Sect. Aptera Kusnez.Roburic acid, a pentacyclic triperpenoid, which has a high content in G. straminea Maxim., was recorded to be the standard substance of Radix Gentianae Stramienae in the Chinese Pharmacopoeia (2010 edition). Meanwhile, We established a method for the purification of roburic acid from the petroleum ether crude extract of G. straminea Maxim. by HSCCC coupled with ELSD, which showed to be easy, accurate and can be used in the large-scale preparation of standard substance of ruburic acid.
