Objective The thesis aims to investigate the expression and distribution of Caspase-8,RIP and nuclear transcription factor-κB(NF-κB) in oral lichen planus(OLP),and analyze their relationship with the events of cell apoptosis in formation and progression of OLP,further reveal the origin of disease formation and offer theoretical and experimental foundation for developing new treatment.Methods Immunohistochemical technique with SP method was used to detect the expressions of Caspase-8,RIP and NF-κB in 30 OLP cases and 15 normal oral mucosa specimens.TUNEL was used for detecting the cell apoptotic index(AI) in 15 OLP cases and 5 normal oral mucosa specimens.The experimental data is subjected to statistical analysis.Results Compared with the control group,the increased AI of epithelial cells(6.76±2.32) and the decreased AI of lymphocytes(1.75±0.74) were found in OLP(P=0.000<0.05).The positive rate of Caspase-8,RIP and NF-κB of epithelial cells were 96.67%,86.67%,93.33%respectively, remarkably higher in OLP than in normal control(P<0.05).The positive rate of Caspase-8,RIP and NF-κB of lymphocytes were 100%,90%,80%respectively,significantly higher in OLP than in normal control(P<0.05).A positive correlation was observed between Caspase-8,RIP and NF-κB expression of epithelial cells and AI of itself;the expression of Caspase-8,RIP and NF-κB of lymphocytes was found positive related to AI of itself,a positive correlation was also observed between NF-κB expression of lymphocytes and AI of epithelial cells.There was a positive correlation between the expression of Caspase-8,RIP,NF-κB of epithelial cells and lymphocytes in OLP.Conclusion(1) There is a significant difference of cell apoptotic index(AI) between epithelial cells and lymphocytes in lamina propria in OLP.Compared with the normal group,OLP is found increased in AI of epithelial cells and decreased AI of lymphocytes,which indicates that the abnormal cell apoptosis plays an important role in the pathogenesis of OLP.(2) Over expression of Caspase-8,RIP and NF-κB in epithelial cells and lymphocytes in lamina propria is positive related to AI of itself,a positive correlation was also observed between NF-κB expression of lymphocytes and AI of epithelial cells,it is suggested that over expression of Caspase-8,RIP and NF-κB in OLP may play a partial role in the pathogenesis of OLP.(3)There is a positive correlation between the expression of Caspase-8,RIP and NF-κB in OLP,which suggest that Caspase-8,RIP and NF-κB are involved in the pathogenesis of OLP.(4) Abnormal cell apoptosis may play an important role in the pathogenesis of OLP,furthermore,Caspase-8,RIP and NF-κB have participated in the abnormal cell apoptosis of adjustment.
