| Aims:①Use the animal to do experiment,discover the mechanism of healing effect of disease of knee joint synovitis.②Observe the molecule mechanism of curative effect of Tra-Ch-medicine(the xibizhuang sour) for curing knee joint synovitis.Methods:12 white male rabbits about three or four months,about 2.2-2.8Kg weight are divided them into two groups in the rule of equilibrium:named model group and Tra-Ch-medicine group,amongst model group,the normal legs belong to comparison group,18 joints.Concrete Methods of the knee joint synovitis animal model:Fix the white rabbit,remove the rabbit' hair by Na2S(8%) and disclose the skin.Inject sodium pentobarbital(1ml/kg,3%),Use acupuncture needle to penetrate into the middle of leg quadriceps and calf-intestines muscle,connect with acupuncture diagnose equipment,then electrify the equipment and choose continuous waveform about 50Hz frequency,6.0mv intension In order to set up knee joint synovitis model,rabbits' legs must be bending and extending ceaselessly,once everyday,90min every time,10 days together, which will led to knee joint synovitis.From the first day of making the models,Tra-Ch-medicine group begins to force the rabbit to take medicine (Every 1ml liquid medicine contains 1g unprocessed drug).Rabbit's measure with the medicine is about 4.7ml/kg.Once everyday and 10 days' treatment.The model group gives rabbits 0.9%equal quantity of physiological saline,measures the perimeter of the knee joint and takes notes.After eleven days,put the rabbits to death by tying their necks with the air stuff,then cut open the bag of knee joint.After fundament observation and note-take,use 1ml physiological saline to penetrate and wash the cavity of joint,collect the liquid of joint.Then put it in the silicon test tube which has already molten and refrigerate(4℃,1800r,10min) after observe the situation of its chromatic ness and take notes with naked eyes.The collection of clear liquid should be kept in cold storage at-70℃and kept stand-by.Result:(1)Compared with model group and comparison group,the knee joins of the model group are swollen and the Synovitis become thick.At the same time the content of Interleukin-1β(IL-1β)(9.22±2.30pg/ml,170.76±38.00pg/ml) and Tumor Necrosis Factor-α(TNF-α)(5.40±0.40pg/ml, 55.45±9.39pg/ml) are heightened remarkably.After the statistics of scripture does with it,and it has remarkable difference(P＜0.05).(2)Compared with model group and medicine group,the knee joins of the model group are swollen a bit and the Synovitis become thick a bit.At the same time the content of IL-1β(9.22±2.30pg/ml,170.76±38.00pg/ml)and TNF-α(6.54±0.63pg/ml,92.20±28.29pg/ml) are increased remarkably a bit,After the statistics of scripture does with it,and it has remarkable difference(P＜0.05).Conclusion:①IL-1βand TNF-αare relate to the process of knee joint synovitis closely.②The xibizhuang medicine can low the content of IL-1βand TNF-αin the liquid of knee joint,restrain the production of knee joint synovitis,reveal that the mechanism of healing effect of disease that the traditional Chinese medicine on the level of molecule cures the knee joint synovitis.
