Expression Of TTF-1 And Thyroid Specific Protein In The Development Of Rat Embryo

ObjectiveTo Observe the expression of thyroid transcription factor -1(TTF-1),thyroglobulin (TG) and thyroid peroxidase(TPO) during embryonic development in rats,and to evaluate the relationship among TTF-1 and TG,TPO and thyroid differentiation.MethodsUsing the method that is checking off vaginal suppository to determine whether the rat have pregnancied.Using the HE stain staining under optical microscope in search of thyroid in the embryonic days of 7,9,11,13, 15,17,19.Using immunohistochemistry techniques and the image analysis system(image pro plus,IPP) to analysis.Using x±s to express measurement data,and using linear and multiple linear correlation analysis;Using LSD-t (least significant difference-t) test to contrast between two groups.Results1.There is no thyroid that could be observed in the embryonic day of 7,9,11. TIT-1,TG and TPO were not expressed there.2,TrF-1,TG and TPO were all expressed in the embryonic day of 13,15,17 and19,mainly located in the cytoplasm.They were correlated with embryonic day. The r were 0.614,0.661 and 0.641,all of their p＜0.01.TIF-1 was correlated with TG and TPO,The r were 0.309 and 0.406,both of their p＜0.01;TPO was correlated with TG,The r was 0.466,p＜0.01.3.Thyroid differentiation was correlated with age,the R was 0.932,p＜0.01,and also with TTF-1,the R was 0.644,p＜0.01.Conclusions1,There are gradual growth trend about TTF-1,TPO,TG which expressed in rat embryo with the embryonic day.TTF-1 impacts the expression of TPO and TG. 2,TTF-1 also can promote thvroid ceils to differentiation.3,The expression of TPO and TG was correlated.
