Caragana opulens Kom.,a species of Caragana genus in the family Fabaceae, is a chilling sensitive shrub growing in some Chinese semiarid areas:Inner Mongolia, Shanxi,Ningxia,Shaanxi,Gansu,Qinghai,Tibet,Sichuan,and etc.C.opuIens of which Tibetan name is Cha Ma,has a profound medicinal history in Tibetan traditional medicine.Cha Ma taste bitter and is cool-natured,can cure all kinds of fever,vein disease and has an effect of emetic.we never saw any report about the chemical composition and pharmacological activity of C.opulens so far.In this thesis,we report the systematic studies of its chemical constituents,and measured the anti-acetylcholinesterase activity of the compounds.Ethanol extract was extracted by petroleum ether,chloroform,ethyl acetate,n-butanol successively and systematic isolation of Petroleum ether,ethyl acetate,chloroform extract of C. opulens was carried out.Ten compounds were obtained by means of repeated silica gel column chromatography,Sephadex LH-20,MCI column chromatography,TLC,recrystalization and Preparative TLC et al.Based on the modern spectroscopic methods(1HNMR,13CNMR,2D-NMR,EI-MS,TOF-MS, IR,etc.),those compounds were identified as:2H-benzo[g]indol-2-one(1), 9,12,13-Trihydroxy-10(E)-octadecenoic acid(2),p-hydroxy-acetophenone(3), vanillic acid(4),p-hydroxy-trans-cinnamic acid(5),trans-cinnamic acid(6), 3-hydroxy benzoic acid(7),glycerol monoester heptacosane(8),lupeol(9),β-sitosterol(10).1 is a new compound while compounds 2,3,4 are isolated from genus Caragana for the first time.We carried out TLC-bioautography test to screen compounds which have anti-acetylcholinesterase activity,found that compounds 3(10μg) has obvious inhibitors spots in the TLC plate,the activity were further confirmed by Ellman method and found that it had a week inhibition on acetylcholinesterase activity,where 50%concentration of inhibition(IC50) was 1.43 mM.This paper not only have a certain understanding for the chemical constituents and anti-acetylcholinesterase activity of Caragana opulens Kom.,but also provide a scientific basis for further research and development on Caragana resources.
