Caragana microphylla Lam belongs to the genus Caragana of leguminous plants. This plant grow in shrub, it is 1 to 2 meters high, distributed in shanxi, gansu, sichuan, anhui, Tibet, northeast and other place in China. Caragana microphylla Lam resistant to drought, cold, and high temperature. It is one of the most suitable tree species which build forest of firewood forests, forage, shelter. Caragana microphylla is called bone grass by Korean medical. It is commonly used as folk medicinal materials. Its medicinal part is root which tastes bitter, with promoting blood circulation, removing blood stasis, expelling wind damp, diminishing inflammation, cardiotonic effect, terminal sedation, diuresis effect an so on. It is used in the treatment of osteomyelitis, rheumatoid arthritis, high blood pressure, gynecopathy, etc. Experiments showed that the root of Caragana microphylla ethyl acetate and n-butanol extraction possessed antitumor activity. In order to discover the compounds with anticancer activity, it is necessary that its activesite were separated, and explore the resources of medicinal plants.The root of Caragana microphylla was extracted with 95% ethyl alcohol by heating reflux solvent. The ethanol extract is suspended in water and extracted with petroleum ether, ethyl acetate in turn, finally remaining water extraction. The extraction of ethyl acetate and water was separated and purification by silica gel column chromatography, D101 macroporous adsorption resin, reversed-phase silica gel column chromatogramphy,SephadexLH-20.By physical and chemical property and spectral data, there were six compounds isolated and identified from the extraction of ethyl acetate,they were lup-20(29)-en-3-one (1)、maackiain (2)、formononetin (3、 syringaresinol (4)、calycosin (5)、lariciresinol (6),two compounds were isolated and identified from the extraction of water as liriodendrin (7)、hypaphorine (8).Compound (1)、(4)、(5)、(7) and (8) were isolated from Caragana microphylla for the first time. In vitro cytotoxicity experiment, the result showed that compound 5 had toxic effects to the hepatoma cell line Hep3B.According to the report, compound 3、6 have anticancer activity.
