| The apoptosis model of spleen lymphocytes of mice was established by dexamethasone,the inhibition of Ganoderma lucidum Spores Powder on apoptosis of spleen lymphocytes of mice were studied through detecting the index of immune organs,the MDA content in serum and the apoptosis of lymphocytes with flow cytometry and TUNEL(Terminal deoxynucleotidyl transferase dUTP nick end labeling).The determination results of immune organs index indicated that each group of Ganoderma lucidum spores powder could inhibit the decline of immune organs index induced by dexamethasone at different degrees,among which the high dose group of Ganoderma lucidum spores powder had the best effect.The determination results of MDA content in serum suggested that the high dose group of Ganoderma lucidum spores powder could prevent the increasing of MDA content in serum induced by dexamethasone (p<0.05),and inhibit the dexamethasone-induced lipid peroxidation.The detection results of flow cytometry indicated that the apoptosis of spleen lymphocytes in model group were very obvious,the difference between model group and groups of Ganoderma lucidum spores powder were quite significant;All dose groups could inhibit the dexamethasone-induced apoptosis of spleen lymphocytes,among which there are dose-dependent relationship, and the high dose group of Ganoderma lucidum spores powder had the best effect(p <0.01),with its apoptosis close to the control group.The results of TUNEL staining revealed that a large number of TUNEL positive cells presented in the spleen of model group,while only a small number of TUNEL-positive cells in the high dose group of Ganoderma lucidum spores powder. The statistical results of Apoptotic index of each group indicate that all dose groups of Ganoderma lucidum spores powder differed significantly from the model group,among which there were dose-dependent relationship,and the high dose group of Ganoderma lucidum spores powder had the best effect(p <0.01).This result coincided with the results of flow cytometry detection.Based on the results above,we concluded that Ganoderma lucidum spores powder can significantly prevent the decline of immune organs index of mice induced by dexamethasone, reduce the MDA content, inhibit dexamethasone-induced apoptosis of spleen lymphocytes significantly. |
