Experiment Study In Treatment Of Adolescent Polycystic Ovary Syndrome By "Neizhang" Pill

Objective:The aim of this study was to investigate the effects of Neizhang Pill in immunology,especially the expression of tumour necrosis factor-alpha(TNF-α) and Interleukin-6(IL-6) in blood serum,of rats with polycystic ovary by light-microscope,HE dyeing and immunohistochemical method,in order to explore the possible mechanism of Neizhang Pill in cells and molecules levels and direct the application in clinic.Material and method:24-day-old immature female SD rats(n=60) were separated into two groups at random.One group was PCOS animal model(n=50),the other group was used as control(n=10).Progesterone-synchronized immature rats were used as the animal model for polycystic ovary(PCO) by injecting 1.5 IU HCG twince daily to 36 days old.And then separated 3 groups(10 rats per group),the model group treated with Neizhang Pill for 30 days,separated 10 rats from the model group as the control group.After treatment,the rats were killed.Blood specimens were centrifuged at 3000r/min for 20 minutes,the supernatant was collected for measuring TNF-α,IL-6.Statistics:All experimental data were analyzed by means of Student's t-test,analysis of variance,Newman-Keuls test or linear correlation and processed by SPSS 14.0.There was a statistical significance when P<0.05.Results:There were no difference of body weight between Neizang Pill treatment groups and control group before pouring Neizhang Pill.But after pouring,the treatmentgroups significantly lightened.In PCO rats the number of Iuteum decreased but the number of cystic follicles increased contrarily.The cellular layer of ovary granulosa cell of the rat with polycystic ovary got thinner and thecal cell became thicker.Neizhang Pill had the effect of increasing the cellular layer of ovary granulosa cell and decreasing the cellular layer of thecal cell for the rats with polycystic ovary.In PCOS group(36days),serum level of tumour necrosis factor-alpha was signifieantly higher than this in control group(P<0.01);After pouring,in the high and middle treatment groups TNF-αwas remarkably lower than this in the PCOS group(P<0.01);But in the low treatment group TNF-αwas lower than this in the PCOS group(P<0.05);The high treatment group was no differ from the control group(P＞0.05).In PCOS group(36days),serum level of IL-6 was higher than this in control group(P＜0.01);After pouring,in the high and middle treatmentgroups IL-6 was significantly higher than this in the PCOS group(P＜0.01);In the low treatment group IL-6 was lower than this in the PCOS group(P＜0.05).Conclusions:This study set up PCO rat modle,the changement of ovarian pathology and immunocell factor are similar as the patient of PCOS.Neizhang Pill could effect on PCO rats' ovarian morphology,and by this way to improve ovarian function.Immunocell factor IL-6 and TNF-αencoded inflammation steped up in the serum of PCO rats,which cued that PCOS related closely with immunology and PCOS maybe a chronic deuto-clinic inflammation.Immunocell factor IL-6 and TNF-αencoded inflammation were degraded by Nizhang Pill,which cued that immuno-internal was improved by Nizhang Pill.
