ObjectiveTo compare the mRNA expression levels of survivin,hTERT,STK15 between laryngeal squamous cell carcinoma(LSCC) and corresponding normal tissue and to examine their protein expressions.Then to seek the relationships between their mRNA expression levels and clinical characteristics(gender of patients,age of patients,clinical status and cervical lymph node metastasis) respectively.Finally,to analyze the correlations of their mRNA expression levels in LSCC.The subject is performed to find new genes for the early diagnosis of LSCC and to develop new ideas for the genetic therapy of LSCC.MethodsThe mRNA expression levels of survivin,hTERT,STK15 in 35 LSCC and corresponding normal tissue were detected by real-time PCR.The protein expression levels of survivin,hTERT,STK15 in 35 LSCC and corresponding normal tissue were tested by Western-blot.Statistical analysis was performed by SPSS13.0.The differences of the mRNA expression levels of survivin,hTERT,STK15 between LSCC and corresponding normal tissue were analyzed with paired samples t-test.The relationships between the mRNA expression levels of survivin,hTERT,STK15 and the clinical characteristics were analyzed with independent samples t-test.The correlations of their mRNA expression levels in LSCC were analyzed with Pearson correlations.P<0.05 is regarded as the difference has statistical significance.Results1.The mRNA expression levels of survivin,hTERT and STK15 were significantly up-regulated in LSCC than those in normal tissue(survivin:t=6.91,P<0.01;hTERT: t=6.83,P<0.01;STK15:t=6.85,P<0.01).2.The protein expressions of survivin,hTERT,STK15 were significantly higher in tumors than those in corresponding normal tissue.3.No statistically significant relationships of their mRNA expression levels were observed between the patients with different gender,age,clinical status and cervical lymph node metastasis.4.The expressions of survivin,hTERT,STK15 mRNA were positively correlated with each other,survivin-hTERT:r=0.633,P<0.05;survivin-STK15:r=0.627,P<0.05;hTERT-STK15:r=0.784,P<0.05.ConclusionBoth mRNA and protein expression levels of survivin,hTERT and STK15 were significantly up-regulated in LSCC,which suggests that all of them are involved in the occurrence of LSCC,even they may cooperate with each other during this process. Thus,survivin,hTERT and STK15 may become the new indexes for the early diagnosis of LSCC and the new targets for genetic therapy.
