| ObjectiveTo study the effects of KI on the proliferation activity of human skin fibroblasts at different doses,based on general principles of cell culture,in order to exploring the new uses for iodine,guiding clinical treatment.Methods1.Cellular morphologies were observed by inverted phase contrast microscope on the proliferation activity of human skin fibroblasts at different doses(0,1000,3000,5000,7000,9000,11000μg/L) after 24 hours,and Cell growth and functional activities were studied.2.Flow cytometry(FCM) was used to determine and compare the cell cycle of the different fibroblasts.Data was collected and expressed as mean±standard error of mean.The significance of any differences was evaluated by student's t-test.The level of statistical significance was set at P<0.05.3.The methods of ELISA were used to detect the proliferation activity of human skin fibroblast VEGF and IGF-1 concentration were measured and analyzed.Results1.The isolated fibroblasts could grow and proliferate in vitro,and the culture cells were fusiform mononucleate cells with typical ultrastructure of fibroblast under TEM at certain range of concentration(<5000μg/L).Human skin fibroblasts were the atypical morphological characteristics of fibroblasts at certain range of concentration(>7000μg/L).2.The FCM results showed that concentration Iodine could significantly increase S phase proportion of fibroblasts at certain range of concentration(<5000μg/L);but decrease S phase proportion of fibroblasts at certain range of concentration(>7000μg/L),The S phase proportion of fibroblasts was highest from 5000μg/L to 7000μg/L,the differences were significant compared with the iodine free control.3..concentration Iodine could significantly increase the proliferation activity of human skin fibroblast at certain range of concentration(< 5000μg/L),but decrease the proliferation activity at certain range of concentration(>7000μg/L).The proliferation activity was highest from 5000μg/L to 7000μg/L,the differences were significant compared with the iodine free control.Conclusion:1.Iodine,the exposure level is<5000μg/L,can increase the proliferation activity of human skin fibroblast,whereas decrease the activity as the exposure is>7000μg/L.2.Appropriate low concentration of iodine can play a important role on the proliferation of fibroblast.Appropriate low concentration of iodine has important theoretical and practical significance in exploring the new uses for iodine,guiding clinical treatment,and increasing patient safety in taking medicine.3.Befitting iodine could promote the expression of VEGF and IGF-1 in human skin fibroblast accordingly,promote wound healing. |
