Objective:To investigate the distribution of LIPC gene-250G/A polymorphism in Han and Dongxiang population in Gansu province. And to investigate the association between LIPC gene -250G/A polymorphism and type 2 diabetes mellitus in Han and Dongxiang population in Gansu province.Methods:A total of 428 unrelated subjects in Gansu, including 205 Dongxiang population(98 type 2 diabetes and 107 normal glucose tolerant (NGT) controls), and 223 Han population (102 T2DM and 121 NGT), were recruited. Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) were employed to screen the polymorphism(-250G/A) in promoter region of LIPC gene. Biochemical indicator and phenotypes related to glucose and lipid metabolism were measured. Excel was used to build database, SPSS 10. 0 was used for analyses.Results:1. Patients with T2DM were correlated with central obesity, insulin resistance, hypertension and dyslipidemia in Han and Dongxiang population in Gansu province.2. In Dongxiang population, -250G/A genotype frequencies of GG,GA,AA were 0.4486,0.3678;0.4579,0.5112; and 0.0936,0.1225; in NGT group and T2DM group respectively and allele frequencies of G, A were 0.6776,0.6225 and 0. 3224, 0. 3775, in NGT group and T2DM group respectively; There were no significant differences in genotype and frequencies of allele in LIPC polymorphism between NGT group and T2DM group (P>0. 05). 3. In Han population, -250G/A genotype frequencies of GG, GA, AA were 0.4959,0.4216; 0.4298,0.4608 and 0.0744, 0.1176, in NGT group and T2DM group respectively and allele frequencies of G, A were 0.7107,0.6520 and 0.2893,0.3480, in NGT group and T2DM group respectively; There were no significant differences in genotype and frequencies of allele in LIPC polymorphism between NGT group and T2DM group (P>0.05).4.Anthropometric and clinical features comparison in Dongxiang population. In NGT group, the subjects with AA genotype had the significant increased HOMA-IR compared with those who carried GG genotype ( P <0.05) ; In T2DM group, the subjects with AA genotype had the significant increased levels of DBP compared with those who carried GG genotype ( P<0.05).5. Anthropometric and clinical features comparison between NGT group and T2DM group in Han population. In NGT group, the subjects with AA genotype had the significant increased levels of Fins, TG and HOMA-IR compared with those who carried GG genotype( P<0.05) ; In T2DM group, the subjects with AA genotype had the significant increased levels of TG compared with those who carried GG genotype (P<0.05).6. The frequencies of genotype and allele of-250G/A polymorphism in the LIPC gene in Dongxiang was higher than those in Han; But it had no significant differences (P>0.05).Conclusion:The distribution of-250G/A polymorphism in the LIPC gene appears to be no difference between Han and Dongxiang population. the G-250A promoter variant of the LIPC gene is associated with insulin resistance in Han and Dongxiang population and may contribute to increasing the levels of TG in Han population.
