Expression And Clinical Significance Of ADAM-12 And PCNA In Bladder Carcinoma

Objectives : To investigate the expression of adamalysin 12 (ADAM 12) and PCNA in human bladder carcinoma and to explore the correlation of ADAM 12 and PCNA in different grades and stages of bladder cancer.Methods : Biopsies cases of 15 normal bladder issues and 43 bladder tumors cases were analyzed. And immunohistochemistry was conducted to detect the expression of ADAM12 and PCNA in the biopsies in the method of streptavidin avidin - peroxidase (sp). We analyzed the relationship between bladder transitional cell cancer of stage and pathological grade with their expression rate. And to find out whether there was correlation in the expression of ADAM 12 and PCNA in bladder cancers.Results: There were 43 cases tissue of bladder transitional cell carcinoma patients and 15 normal bladder tissue . Postive expression signals of ADAM12 were detected significantly higher in the bladder cancer biopsies than those in the normal ones ( P=0.0105). Those with higher histological stage had a higher expression level of ADAM-12, compared to the lower histological stage (P <0.0001). And those with higher histological grade had a higher expression level of ADAM-12, compared to the lower histological grade (P=0.005).Expression rate in patients with high degree of differentiation is lower than low degree of differentiation. Positive expression rate of PCNA were detected significantly higher in the bladder cancer biopsies than those in the normal ones ( P=0.0261). Those with higher histological stage had a higher expression level of PCNA (P = 0. 0137). And those with higher histological grade had a higher expression level of PCNA (P = 0.0043). Meaning time, we find there is positive relationship between the expression of ADAM-12 and PCNA in bladder cancer in stages and grades, (r= 1.000, P<0.0001).Conclusions: The overexpression of ADAM12 and PCNA in the biopsies of bladder rumors shows that protein expression of ADAM-12 and PCNA correlated with tumor stage and grade. And in our study those with higher histological stage and grade had a higher expression level of ADAM-12 and PCNA compared to the lower histological stage.ADAM-12 and PCNA, in particular ADAM-12 is the potential value of bladder cancer tumor marker, is expected to become a new indicator of clinical diagnosis and prognosis. When use the two indicators together is likely to increase the detection efficiency.
