| Objective:Cervical cancer is most common woman genital system malignant tumor second only breast cancer, and showing young tendency. Although the incidence of cervical cancer has decreased notably, the long term survival rate is lower. For sake of cutting down the incidence and mortality, it is critical for screening precancerous lesion and detecting prognosis. The purpose of this study is to detect the expression of the piwil2 in normal cervix ,cervical intraepithelial neoplasia, cervical cancer and Hela cell that before and after treatment,and then discuss the clinical significance of piwil2 in cervical cancer.Methods:1) To investigate the expression of piwil2 in normal cervix ,cervical intraepithelial neoplasia, cervical cancer, the Western Blot were used.2) IHC was used to examine the expression of piwil2 in normal cervix ,cervical intraepithelial neoplasia, cervical cancer.3) Western blot were used to examine the expression of piwil2 in Hela cell that before and after treatment.4) IHC were used to examine the expression of piwil2 in Hela cell that before and after treatment.5) To detected the expression of piwil2 in cervical cancer patients of differente pathological types. Results:1) The expression of piwil2 in cervical intraepithelial neoplasia, cervical cancer is higher than that in normal cervix , P﹤0.05.2) The expression of piwil2 in Hela cell that after treatment is lower than the control group, P﹤0.05.3) There is no significant difference in the serum level of piwil2 between cervical cancer patients of differente pathological types, P﹥0.05.Conclusion:The positive expression and amplification tendency of piwil2 have presented upgrade in uterine cervix with the advancement of pathological changes. Compared with normal cervix, there was a significantly difference in cervical intraepithelial neoplasia and cervical cancer. The expression of piwil2 in cervical cancer have no correlation with pathological types. The over expression of piwil2 may play an important role in cervical carcinogenesis, the piwil2 gene of pathological changes could be used as a better tumor index for early cervical cancer screening and predicate the the advancement and evaluate poor prognosis of cervical leision; meanwhile it decrease obviously in Hela cell after treatment,so it also could be used as a better index for treatment effect. |
