ObjectiveThe gastric carcinoma is common and frequently-occurring disease in China. Identification of the key molecular targets involved in gastric carcinoma has significant therapeutic implications.However,the genetic and epigenetic events of gastric carcinoma are poorly understood.Twist,with the characteristics of a cancer gene,can inhibit apoptosis through variety ways, and also plays an important role in the process of tumor formation, development and transfer.To investigate the expression and clinical significance of transcription factor Twist in human gastric carcinoma.MethodsImmunohistochemistry(SABC) was used to detect the expression of the Twist protein in 50 cases of human gastric carcinoma and 50 cases of paraneoplastic tissue.Twist mRNA was detected in 50 cases of gastric carcinoma and 50 cases of paraneoplastic tissues by real-time quantitative PCR.Results1.Immunohistochemistry showed that in the gastric carcinoma and paraneoplastic tissues,the positive rate of Twist protein were 72%and 14%, the expression of Twist in gastric cancer tissues was much more higher than the paraneoplastic tissue(P =0.000).Twist protein expression in gastric carcinoma in different sex,age,tumor sites has no significant difference(P>0.05);but in cancer differentiation,depth of invasion,lymph node metastasis, TNM stages and the situation of distant metastasis,the expression of Twist protein shows significant difference(P<0.05).2.Real Time-qPCR showed that the quantity of the Twist mRNA in cancer tissue was higher than the paraneoplastic tissue(P=0.000).Twist mRNA expression in gastric carcinoma in different sex,age,tumor sites has no significant difference(P>0.05);in cancer differentiation,depth of invasion, lymph node metastasis,TNM stages and the situation of distant metastasis, the expression of Twist mRNA shows significant difference(P<0.05).3.The Twist expression was not significant difference in different group of age and the patients' gender,while it was significant different in the group of infiltrative depth,lymphatic and distant metastasis,and histological grade.Conclusions1.The expression level of Twist protein and Twist mRNA in gastric cancer tissue were significantly raised than the adjacent normal tissue,suggested Twist gene might be involved in the formation of gastric cancer.Take effective control measures may have better results in the cancer prevention and control.2.The expression of Twist protein and Twist mRNA had a significant difference in different TNM stage,the situation of lymph node metastasis, tumor differentiation and the distant metastasis,and there was positive correlation between them.That suggested Twist gene may play an important role in the development,invasion and metastasis of gastric cancer,if we have an effective intervention in the stage of cancer progression,we can expect a better treatment effect.
