Objective: To observe the changes of cytosol-free calcium concentration in smooth muscle in partial unilateral ureteral obstruction rats.Method: Eighty Wistar rats had been divided into four groups by random: 8- week experimental group (n=20) and sham control group (n=20), 16- week experimental group (n=20) and sham control group (n=20). Experimental animals left ureters (n=40) had been embedded the upper half into the psoas muscles. Sham control rats only received laparotomy After the model arc established the rats are separated in different experimental point to carry out the intracellular calcium concentration were measured in single ureteral smooth muscle cell in rats using the fluorescent calcium indicator dye Fura-2/AM at rest situation.,and oberve the change of ultra microstructure of mitochondria with electron microscope.Results: the intracellular calcium concentration in single ureteral smooth muscle cell in PUUO rats had no significant difference in the initial stage at rest situation in comparison to sham control group(200.42±36.34 vs 187.63±23.51 ).However,at 16th week after onest, the intracellular calcium concentration in single ureteral smooth muscle cell(454.92±31.69)was significantly increased in PUUO rats as compared with that in sham control group(173.88±35.44).Conclusion: intracellular calcium overload occurred later than the dysbolism .Maybe intracellular calcium overload is an important factor in the progression of PUUO.
