ObjectiveTo investigate the influence of acute urinary retention (AUR) duration on the function and structure of the kidney in rats.Materials and MethodsAUR was induced in Sprague-Dawley rats by forced saline infusion. Forty rats were divided into five groups and subjected to AUR for 1 hour(h),2h,4h or 8 h; control rats were subjected to a sham operation without AUR. Renal function was assessed by analysis of serum creatinine (SCr) and blood urea nitrogen (BUN). The haemodynamics of the renal arteries was observed by ultrasonography; Changes in renal structure were examined by terminal deoxynucleotidyl transferase (TdT)-mediated uridine triphosphate nick end-labelling (TUNEL) and transmission electron microscopy (TEM).ResultsCompared to controls, rats subjected to 2h,4h or 8h of AUR showed significant, time-dependent increases in SCr, BUN, renal size and renal pelvis separation (P< 0.05). However, the thickness of the renal cortex did not change significantly with increasing duration of AUR. Four or eight hours of AUR groups elicited time-dependent increases in blood flow rate of the main renal artery (MRA), the interlobar renal artery (IRA) and the segmental renal artery (SRA; P< 0.05). The resistive index (RI) of the MRA, IRA, and SRA also increased significantly with increasing duration of AUR. Prolonged AUR increased the apoptotic index of the renal tissues by TUNEL and resulted in more severe renal injuries.ConclusionsIncreasing AUR duration induces more severe injuries of renal function and structure in rats.
