Objective:The purpose of this study was to find out the best prescription method for Yantongping transdermal ointments through Orthogonal Design and research on its function of Anti-inflammatory and Analgesis and Invigorating blood circulation.Methods:With 60 minutes in vitro cumulative penetration rate, using L9(34) orthogonal experiment, find out the best formula and technology through grinding method, and evaluate its quality thereafter. picked up 36 male mice weight for 19±1 g, randomly divided into six groups, led them with ankle inflammation by Formaldehyde, then plastered them with 0.1 g. (cm2)-1 Vaseline,0.1 g. (cm2)-1Vaseline and Fatty Alcohol,0.1 g. (cm2)-1 Yantongping transdermal ointments (low dosage),0.3 g. (2cm2)-1Yantongping transdermal ointments (medium dosage),0.5 g. (3cm2)-1 Yantongping transdermal ointments (high dosage), 0.1 g. (cm2)-1 Voltaren Emulsions with the aim to compare the function of Anti-inflammatory of each medicine by observing Ankle perimeters in different time periods of the animals before and after inflammation; in the second group, picked up 36 female mice weight for 19±1 g, randomly divided into six groups as above, compared the pain threshold and its the percentage increase of mice by the methods of Woolfe hot board,aiming to observe the difference of abirritation of each medicine in different time periods; in the third group, picked up 36 mice (18 male and 18 female) by randomly divided into six groups as above, compared of the number of body torsion and inhibition rate of the mice after each medicine's plaster by methods of mouse writhing response, aiming to observe the difference of abirritation of each medicine in different time periods; in the fourth group, picked up 72 mice (36 male, 36 female), divided into 2 groups as the blank group, or the model group, each divided into six smaller groups as above, measuring△A,△V before and after modeling by Microcirculation method and Microscopic micro-distance measuring method, while compare the blood circulation function of each medicine by the scoring of the blood of veinule.Results:The best prescription is confirmed by A2B2C3D2, that is with the weight of the traditional Chinese drugs (TCD) vaseline ratioes (A) to 50%, with the weight of TCD fatty alcohol ratioes (B) to 25%, the order of reagent is as follow:fatty alcohol, TCD, vaseline, the grinding temperature is 80℃, and according to six quality evaluation as physical appearance, melting point, water value, stability, irritation, got the results that the differences between the group of Yantongping (low dosage) in 2 h,4 h, (medium dosage) in 1 h, (high dosage) in 0.5 h and vaseline had statistical significance, (significantly less than vaseline group) (P<0.05); in the Yantongping group (medium dosage) in 1.5h,2h,4h,6h, the Yantongping group (high dosage) in 1h,1.5h,2h,4h,6h had obvious statistical significance over vaseline group in△L level, (obviously less than vaseline group) (P<0.01). The pain threshold and the percentage increase pain threshold level measured showed that the Yantongping group (low dosage) has no statistical significance over vaseline group in different time periods (P>0.05); the Yantongping group (medium dosage) in 60min and the Yantongping group (high dosage) in all different time periods have had obvious statistical significance over vaseline group, (obviously more than vaseline group). The body torsion and inhibition rate leve measured showed that the Yantongping group (low dosage) has no statistical significance over vaseline group (P>0.05); but the Yantongping group (low dosage) has statistical significance over vaseline group in inhibition ratio (obviously more than vaseline group); the Yantongping group (medium dosage) and the Yantongping group (high dosage) has great statistical significance over vaseline group in the body torsion (obviously less than vaseline group) (P<0.01), in inhibition ratio (obviously less than vaseline group) (P<0.01).Before modeling, in the△A level measured, the Yantongping group (low dosage) has statistical significance over vaseline group in 30 min (significantly more than vaseline group) (P<0.05),while the Yantongping group (medium dosage) and the Yantongpinggroup (high dosage) has obviously statistical significance over vaseline group in different time periods, (obviously more than vaseline group) (P<0.01). In the AV level, the Yantongping group (low dosage) has no statistical significance over vaseline group in all time periods (P>0.05),while the Yantongping group (medium dosage) and the Yantongping group (high dosage) has obviously statistical significance over vaseline group (obviously more than vaseline group) (P<0.01).In the blood flow quality evaluation measured,theYantongping group (low dosage) has statistical significance over vaseline group in 20min,30min (significantly more than vaseline group) (P<0.05); while the Yantongping group (medium dosage) and the Yantongping group (high dosage) has obviously significant statistical significance over vaseline group (obviously more than vaseline group) (P<0.01). After modeling, in the△A level, the Yantongping group (low dosage) has statistical significance over vaseline group in 20min, (significantly more than vaseline group) (P<0.05); the Yantongping group (low dosage) has obviously statistical significance over vaseline group in 30min, the Yantongping group (medium dosage) and the Yantongping group (high dosage) has obviously significent statistical significance over vaseline group (obviously more than vaseline group) (P<0.01). In the AV level measured, the Yantongping group (low dosage) has no statistical significance over vaseline group in different time periods (P>0.05); the Yantongping group (medium dosage) and the Yantongpinggroup (high dosage) has obviously significant statistical significance over vaseline group (obviously more than vaseline group) (P<0.01).In the blood flow quality evaluation measured,the Yantongping group (low dosage, medium dosage,high dosage) has obviously significant statistical significance over vaseline group (obviously more than vaseline group) (P<0.01)Conclusion:Yantongping transdermal ointments prepared the best prescription has a good function of Anti-inflammatory, Analgesis and promoting blood circulation.
