| Objective To investigate the amplification and clinical significance of the TERC gene in cervical lesions, and to determinate the association between the amplification of the TERC gene and the infection of HPV.Methods 100 cases of gynecological admissions for women,Selected in February 2008 to July 2008 Xiangya Hospital, Central South University. The amplification of TERC gene in Cervical exfoliated cells from 21 infiltrating cervical cancer,59 cervical intraepithelial neoplasia and 20 normal were detected by the fluorescent in situ hybridization(FISH). Contemporary, The infection of HPV in 80 cervical disease were detected by the humanpapilloma virus DNA hybrid captureâ…¡(HCâ…¡).Results The positive amplification rates of the TERC gene in normal,CINâ… ,CINâ…¡,CINâ…¢and infiltrating cervical cancer were 0,68.8%.85.0%.100.0% and 90.5%.Compared with the normal or inflammation, the TERC gene positive expression rates in CINâ… ,CINâ…¡,CINâ…¢and invasive cervical cancer were different significantly (P<0.05),and there are significantly difference between CINâ… and CINâ…¢(P<0.05),but there are no significantly difference between the others(P>0.05). The positive amplification rates of the TERC gene in positive and negative of high-risk HPV test result were 90.3%(65/72) and 62.5%(5/8).The TERC gene positive expression rates were different significantly between the two results (P<0.05).The positive expression rates of high-risk HPV in CINâ… ,CINâ…¡. CINâ…¢and invasive cervical cancer were 56.3%(9/16),95.0%(19/20),100.0%(23/23) and 100.0% (21/21).Compared with the CINâ… , the high-risk HPV positive expression rates in CINâ…¡,CINâ…¢and invasive cervical cancer were different significantly (P<0.05),and there are no significantly difference between the others(P>0.05).According to the classification of HPV viral load from low quantity to high. Compared with the CINâ… , the average amount of viral load in CINâ…¡,CINâ…¢and invasive cervical carcinoma was different significantly (P<0.05), and there are no significantly difference between the others(P>0.05)Conclusion Our study suggested that the gain of TERC gene maybe play an important role in cervical carcinogenesis, and it detected by FISH in cervical exfoliated cells is expected to be used as one of the screening method of cervical carcinoma. Genomic integration of oncogenic HPV viral load with cervical lesions increase the severity, HPV DNA testing as cervical intraepithelial neoplasia as a screening method more accurate HPV DNA and gain of the human telomerase gene TERC appear to maybe important genetic events in the progression of cervical lesions. |
