Objective:Through the pre-experimental research, We observed that There are changes of mesenteric, intestinal mucosa, kidney cortex, liver, gastric mucosa, deep fascia, the soft meninges and other micro-circulation in experimental animals, the general voltage electric burns specimens.We can find that not only peripheral tissue microcirculation disturbance but also distant organs were caused by the high-voltage, which caused damage that can be observed that the performance of micro-Yan Bi leukocyte rolling and adhesion, white small thrombus mobility and erythrocyte aggregation phenomena. Many experiments has been verificated that adhesion molecules of selectin family in leukocyte can roll closed to the cell wall of endothelial cells and plays an important role in early adhesion. We designed this experiment aimed to observe the continuous changes of selectin (P, E, L) in voltage electric burn and further explore the mechanism of microcirculation disorder and pentoxifylline on serum selectin expression, These microcirculatory disorders can be prevented and treated for burn injury of high-voltage.This provides a theoretical basis for the possibleilities of reduction mortality and improvement of survival rate.Methods:1. Group:180 healthy grown up male S-D rats are randomly divided into three groups, high-voltage electrical burned experiment group (experiment goup), high-voltage electrical burn PTX reatment group (treatment group) and fake high-voltage electrical burn group (control group),60 rats of each group. The time phase of these groups are divided into six different phases, ahead(T0), immediately(15min) (T1),1h(T2),2h(T3),4h(T4),8h(T5),10 rats of each group. 2. Preparetion:to weigh the rats, number them, The experimental drug concentration required by experiment dubbed.3. Duplication of high-voltage electric burn model:Anaesthetic rats of electrical burn experiment group and PTX treatment group are burned by the 10,000 volts high-voltage electric system for 3 seconds,they are duplicated full-thickness electrical burn with body tissues that touch with the electrodes, we immediately inject PTX(50mg/ml) to rats of treatment group through abdomen, normal sodium to the experiment group and the control group.The progress of control group is the same as the electric burn group,but there isn't electric current in the body of control group rats.4. Collection and detection:In different time phases open pectoral cavity take suction blood about 5ml from rats'heart and fractionate 4ml and lml,the former centrifuge with 3000r/min about 10 minutes,suction supernatant,and then preserve in ultra low temperature freezer.5. The serum content of selectin (P, E, L) of the rats were measure by enzyme-linked immunosorbent assay (ELISA) at different time phases during early electrical burn.6. Analysis of Statistics:all the data was statistically analysis with SPSS16.0 and should be expressed withx-±s.we apply one-factor analysis of variance (ANOVA) among groups.It has significant statistically meaning if p<0.05.Results:the changes of selectin (P, E,L) in the serum1. The serum levels of selectin (P, E, L) haven't obviously change s compared to the control group.(P>0.05)2. Selectin (P, E, L) levelsof early-phase serum of the experimental group higher after the high voltage electric burn and than he high voltage electric burn (P<0.05); in which E-selectin expression level began to increase in an hour after burn from the high-voltage; P-selectin is a high-voltage expression after burn reached a peak and continued in 8 hours; E-selectin expression reached a peak later in 4 hours after injury, and decreased; in different time. The expression of L-selectin after high voltage electric burn reached peak in two hours and decreased gradually with time, but was still higher than that in 8 hours after injury.3. High voltage electric burn after PTX treatment group was not accompanied by an early-phase serum selectin (P, E,L) and electricity before the higher serum selectin (P, E, L) levels (P<0.05), but at the same time compared to electric blow serum selectin (P, E,L) levels decreased to varying degrees (P<0.05); in which E-selectin levels from 1 hour after injury, since high expression; serum selectin (P, E, L) hormone levels in 8-hour-phase electric shock group and treatment groups decreased between the two groups, but no significant difference (P> 0.05).Conclusion:1. selectin (P, E,L) increased after high-voltage electrical burned in experiment groups and PTX treatment groups, maybe play an important role in microcirculatory disorders.2. The study indicates that selectin (P, E, L) was higher in PTX experiment group and than the control group.selectin (P, E,L) might be helpful in prevention and relief microcirculatory disoeder after high-voltage electrical burn.
