Objective: To investigate the long-term effects of high voltage electric burn on platelet activity factor(PAF) and soluble platelet selectin(sP-selectin) in serum of rats and the intervention effects of Ulinastatin and Xuebijing injection.Methods: Healthy clean SD rats were randomly divided into five groups according to the method of digital table: sham injury group,electric injury group,electric injury+saline group,electric injury + Xuebijing group,electric injury+Ulinastatin(UTI) group,70 rats in each group,a total of 350 rats.Each group was divided into seven different time phases according to the same method: 15 minutes before electric injury,1 hour after electric injury,8 hours after electric injury,24 hours after electric injury,48 hours after electric injury,72 hours after electric injury and 1 week after electric injury,with an average of 10 rats in each time phase.Each group was recorded and fed in cages.The experimental high-voltage electric injury system was adjusted to 2KV high-voltage through transformer.The left forelimb and right hind limb of SD rats were used as the ‘entrance' and ‘exit' of electric injury respectively to complete the high-voltage electric injury process.In addition to the sham group and the electric injury group,the rats in the other groups were given intraperitoneal injection of 2ml/kg saline,10ml/kg Xuebijing injection and 2ml/kg UTI respectively.The concentration of PAF and sP-selectin in rat serum was determined by time-phase blood collection(heart blood collection),centrifugation,serum collection and detection of OD value(ELISA double antibody sandwich method).Finally,the data are tested by factorial design variance analysis.Results:1.The concentration of PAF in serum of rats in the electric injury group was significantly higher than that in the sham-electric group(control group)and 15 minutes before the electric injury(all P<0.05),and there were two peaks at 8hours and 48 hours after the electric injury.There was no significant difference between the two groups(P>0.05).After intraperitoneal injection of Xuebijing injection and Ulinastatin,the concentration of PAF in serum of the two groups was significantly lower than that of the electric injury group and the electric injury + saline group(all P<0.05).There was no statistical significance between Xuebijing group and Ulinastatin group(P>0.05),but the decreasing trend of PAF content in serum of rats after drug treatment was observed.Ulinastatin group could inhibit the occurrence of the second peak,and the effect was better than Xuebijing group.2.The concentration of sP-selectin in serum of rats in the electro-injury group was significantly higher than that in the sham-electric group(control group) and 15 minutes before electro-injury(all P<0.05) at 1h,8h,24 h,48h,72 h and 1w after electroinjury,and reached a peak at 8h after electro-injury,and then decreased.There was no significant difference between the electro-injury group and the electro-injury + saline group(P>0.05),that is,there was no significant difference.After intraperitoneal injection of Xuebijing injection and Ulinastatin,the concentration of sP-sselectin in serum of the two groups decreased significantly,which was lower than that of the electro-injury group and the electro-injury+saline group(all P<0.05).Both Xuebijing group and Ulinastatin group decreased the content of sP-selectin in serum,and there was statistical significance between the two groups(P<0.05).Its further mechanism needs to be further explored.Conclusion:1.The levels of PAF and sP-selectin in serum of rats stimulated by high voltage electric burn increased significantly.It is suggested that high voltage electric burn can stimulate the activation of vascular endothelial cells and platelets and release a variety of activating factors and inflammatory mediators,leading to tissue inflammation and coagulation mechanism,affecting the normal physiological function of blood vessels and microcirculation disorders.2.After high voltage electric burn,the concentration of PAF and sP-selectin in serum of rats was significantly decreased by the intervention of Xuebijing and UTI.Therefore,it can be concluded that Xuebijing and UTI have significant effects on inflammatory reaction and circulatory disturbance caused by body injury.
