Objective: Vascular Dementia is the third largest cause of death in our country. Within the last few years,there has been a growing interestin brain injury after Vascular Dementia . Neurotransmitters has been grow downwards after cerebralisehemia, brain cell initiate and execute cell death. Central mediators of apoptosis are one famiIies of molecules:a group of cysteine aspartate proteases,called caspases. Cumulaiive evidence indicates that brain neurotransmitters decrease and apoptosis primarily occurs in the penumbra area,whereas rapid energy depletions inside the ischemic core causes acute necrosis. Semidarkness area has been considered a therapeutic target in cerebral iscbemia. Therefore, it's of great significance to make deeper research into it's pathological mechanism and develop the effective treatment to prevent and cure the VD. Based on the conventional knowledge and the modern research achievements Of Thaditional Chinese medicine(TCM)on VD, my teacher cum Hua Zhuo Jie Du and Huo Xue Hua Yu to cure,clinical has been gain good curative effect.In order to make probe into its tyeatment action mechanism, we adopted VD model of Kunming-mice with the operation of ligating both side carotid artery, observed its effec on mice praxiology, hippocampus path-omorphology, expression of immunohistochermisty caspase-3. the contents of NE,DA in blood serum.Methods: We duplicated the modle of VD mice with the operation of ligating both side carotid arteries. 45 Healthy male Kunming mice were divided into five groups with 9 mice in each group at random, which were sham-operation goup,model group,high dose of Hua Zhuo Jie Du and Huo Xue Hua Yu group(called high-dose group"as follows), low dose of Hua Zhuo Jie Du and Huo Xue Hua Yu group(called low-dose group"as follows), and nimodipine control group (called nimodipine group"as follows). we duplicated the modle of VD mice with the operation of cerebral ischem reperfusion through separating and ligating both side caotid arteries with putting blood from coda except sham-operation group.And then 5 groups were continuously filled medicine orsaline through the stomach from the second day after the operation of cerebral ischem reperfousion. Sham-operation group and modle goup were filled saline and other groups were given medicing. We did the praxiology experiment on the 14th day after operation(jumping standexperiment, water-labyrinth experiment). the mice were cut head for and obtaining Hippocampus tissue, the left part of which was fixed and cut into slices.the tissue was stained by HE(Hematoxylin) . We observed constitutional morphologic changes of Hippocampus tissue under light microscope. To observe the relation of the expression of caspase-3 with Hippocampus tissue,Then we made the blood seru,to part from heart made clear supernatant liquid. We adopt ELISA examined the change of the neurotransmitter (NE and DA) .Results:1 The effect of Hua Zhuo Jie Du and Huo Xue Hua Yu on praxiollogy of VD mice.1.1 The mice's performance record of jumping standEvery period after the operation, modle group had longer reaction time, shorter latency time and more wrong times, there was different (P<0.01) compared with Sham-operation group. At the same time, other groups had shorter reaction time, longer latency time and less wrong times, there was different (P<0.05 or P<0.01) compared with modle group.1.2 The mice's performance record of electricity–labyrinth The day of 14 afteroperation, the wrong times the whole line the modle group were more than those of the sham-operation group (P<0.05 or P<0.01). there was different (P<0.01)compared with the sham-operation group. At the same time, the wrong times of other groups were less than those of the modle group(P<0.05 or P<0.01), and the performance of the High-dose group was better than that of the Low group. There was no significant different between the Little-dose group and the Nimodipine group. 2 The effect of Hua Zhuo Jie Du and Huo Xue Hua Yu on cerebral pathomorphology of VD miceThe pathomorphology under ligh mirror lens indicated that layers of the pyramidal cells in the hippocampus of the modle group were reduced, the pyramidal cells were arranged sparse and missed obviously, the cellnucleuses diminished and the cell structure was vague. Hippocampus of mice brain tissue took on the pathological changes of ischem. Pyramidal cells in the hippocampus of the sham-operation group were arranged closely, cell nucleuses were big and round, nucleoli were clear. The pathological changes of each medical group were improved compared with the modle group.3 consequence of immunohistochemisty about Caspase-3Hua Zhuo Jie Du and Huo Xue Hua Yu High-dose group and Sham-operation group clear that sparing masccline sexual cell. Modle group and Nimodiping group clear that sparing multiplicity caspase-3 masccline sexual cell at all time. caspase-3 masccline sexual cell membrane is very Integrity, cell body to offer oval shap or clostridial form and Cytoplasm offer Buffy drum dyeing. Masccline sexual cell number of Modle groupand Nimodipine group obviously Top medication administration team and Sham-operation group(P<0.05). we could draw the conclusion that there was statistics significance in the difference .4 The effect of Hua Zhuo Jie Du and Huo Xue Hua Yu on the content of NE and DA in the blood serum of the VD mice.The content of NE and DA in the blood serum of the model group were obviously than those of the sham-operation group(P<0.01 or P<0.05);the content of the NE and DA in blood serum of the medical groups were higher obviously than those of model group(P<0.01 or P<0.05);The content of NE,DA in the hippocampus brain tissue of high-dose group was higher than that of the Nimdipine group (P<0.05);we could draw the conclusion that there was different,but there was no statistics significance in the difference between the low-dose group and the Nimdipine group (P > 0.05).Conclusion: 1 The deacend of the study and memory ability resulted from the cerebral ischem is the important reason to cause VD. Hua Zhuo Jie Du and Huo Xue Hua Yu decoction can raise the VD mise's perormence record of electricity-labyrinth and jumping stand experiment and improve the study and memory ability.2 The one of susceptiblest domain with brain tissue is anoxia and ischemia with hippocampus. The damage of the pyramidal cells in the hippocampus tissue is the import pathology.Change that will lead to VD, hot water of to dissolve muddy to neutralize poison and activating blood circulation to dissipate blood stasis have treatment function to the VD mice, and can alleviate the pathology damage caused by the cerebral ischem.3 Neuronal apoptosis plays important role during cerebral ischemia and reperfusion injury, apoptosis to involve in activation Of Caspase-3. Caspase-3 is involved directly in the procedure of neuronal apoptosis. Its expression increases earlier than neuronal apoptosis.4 Hua Zhuo Jie Du and Huo Xue Hua Yu decoction can heighten the content of the NE and DA in the blood serum of VD mice, an alleviate the damage of the cerebral ischem.
