The Effects Of JuanBi Capsules On IL-1,IL-6,TNF-α In Synovial Fluid Of KOA Rabbits After Prevently Administration

Objective: Research Juanbi capsule on knee osteoarthritis rabbit model of preventive medicine,the IL-1,IL-6 and TNF-αcontent of synovial fluid, in order to explore early prevention and treatment to osteoarthritis of Juanbi capsule and its mechanism.Methods: 72 of 4 months Long-eared rabbits,male, body weight (1.80-3.50) kg, divided into six groups randomly, A group (traditional Chinese medicine high-dose prevention group), B group (Chinese herbal medicine dose prevention group), C group (Chinese herbal medicine with low-dose prevention group), D group (glucose-amino acid hydrochloride capsules prevention group), E group (model group), F group (blank control group) each group were 12 rabbits. Besides the control group, each group was used tubular cast immobilization-induced osteoarthritis of rabbit knee model of modeling methods. Immobilized the left knee straight for 6 weeks, so that the left knee is the model of knee osteoarthritis. In modeling time,give the traditional Chinese medicine prevention group with Juanbi capsules, and give the medicine prevention group for glucosamine hydrochloride capsules,model control group was given normal saline for 4 weeks, after 6 weeks of modeling,take rabbits cartilage tissue for gross observation,light microscope observation the same time,joint fluid extraction line ELISA, Across statistical analysis.Results:1. Macroscopic observation, group E articular cartilage degeneration badly, A, B, C, D group to have the varying degeneration, under the microscope observation, except F each group have different levels of cartilage degeneration histological changes. 2. IL-1 of Synovial fluid, middle and western medicine prevention group compared with the model control group were significantly different (P<0.05); medicine high, medium and low-dose groups there was significant difference (P<0.05); Chinese medicine high and middle dose group and the glucosamine capsule was no significant difference (P>0.05).3. IL-6 of Synovial fluid, middle and western medicine prevention group compared with the model group were significantly different (P<0.05); medicine high, medium and low-dose groups there was significant difference (P<0.05); Chinese herbal medicine with the glucosamine capsule dose group was no significant difference (P>0.05).4. TNF-αof Synovial, Western medicine prevention group compared with the model group were significantly different (P<0.05); medicine high, medium and low-dose groups there was significant difference (P<0.05); Chinese herbal medicine with the glucosamine capsule dose group was no significant difference (P>0.05).Conclusion: Juanbi capsule can significantly reduce the IL-1, IL-6 and TNF-αcontent in synovial fluid of KOA in rabbit , enhance cartilage cell metabolism, slow down cartilage degradation, while the play to improve joint function. This is one of the mechanisms for it prevention and control OA.
