Mental Retardation is a kind of disease relevant to intelligence that extend all over the world, serious harm human physically and mentally healthy. Since 1990s later period, the study to non-specific mental retardation genes, especially to X-linked non-specific mental retardation (MRX) genes' searching , location and function study has got great progress. At present, there are 15 genes relevant to MRX has got been ascertained. Though still having many other genes need to be further distinguished, the further study to genes already found on expressing research as well as the relation between expressional level and cognitive ability , is the key question to reveal the relations of these genes with human cognitive ability ,make sure the mechanism of MR. Undoubtly this study also have important medical value and applied prospect.The IL1RAPL1 gene is one of the genes cloned in 1999 relevant to MRX. It locus at Xp22.1-p21.3 of human being X chromosome. We have made sure that IL1RAPL1 gene have high relevant to human senior cognitive ability by the research to the expression of genetic mutation in MRX patient as well as in the animal brain and other organizations. But until mow there are no report about the relations of cognitive ability and the gene's expression level in the brain.This study according to the experimental group and the control group designs. Carry out active avoidance response(AAR) to the adult rat to obtaining different achievements, then use In suit hybridization and Immuno-histochemistry to determine target gene' expression areas in the brain as well as the learning and memory relation with the gene's expression level of mRNA and the protein in the different achievement groups after AAR training. Through comparative analysis, Ascertain the homoerotic gene of target gene' relation with learning and memory .The result show that(l)The rightness rate of AAR does not stabilize within the first 6 days, After the sixth day the achievement' trend going up steadily. The rightness rate of the HALM rats is 93.05% and 27.43% of the LALM in the ninth day.(2) Most Il1raplmRNA locate at hippocampus (Hip),dentate gyrus (DG),Parietal cortex (Pac),peripheral cortex (PRh), piriform cortex (Pir), posterolateral cortical amygdaloid nucleus (PLCo) of the rat brain in which area have deeply correlation with learning and memory. The Il1raplmRNA expression have obvious difference between HALM and LALM(t=6.01, P<0.01), There are no obvious difference between PC and LALM (t=1.19,P>0.05).(3)The Il1irap1 protein mainly locate at hippocampus (Hip), dentate gyrus (DG), peripheral cortex which is relevant to learning and memory which is consistent with Il1raplmRNA distribution.The result of In suit hybridization and Immuno-histochemistry show that Il1raplmRNA and Il1rap1 protein mainly distribute in hippocampus and cortex of the rat brain which have deeply correlation with learning and memory .The result not only have verified the area that the fore researcher' expression about the IL1RAPL1 gene in the brain, but also indicate that the gene may connected with learning and memory.We can further confirm that the gene take part in the learning and memory process from the obvious difference between HALM and LALM of the Il1raplmRNAexpression on in suit hybridization.
