Effects Of Exercise And Zuogui Pills On Biomechanical And Related Hormones In Ovariectomized Rats

PurposeTo explore the effect on biomechanical property and the serum calcitar (CT), bone gla protein (BGP), parathyroid hormone (PTH) and estradiol E2 of the ovariectomized rats by taking both exercise and Zuogui Pills, further to study the therapeutic mechanism.Material and MethodsWistar female rats were randomly divided into 7 groups:normal control group, sham-operation group, model group, estrogen group, aerobic exercise group, Zuogui Pills group and aerobic exercise combined Zuogui Pills group. Aerobic exercise group and aerobic exercise combined Zuogui Pills group had 11 rats, the other groups had 13 rats respectively.Rats in Model group, estrogen group, aerobic exercise group, Zuogui Pills group, as well as aerobic exercise combined Zuogui Pills group received ovariectomy. Rats in sham-operation group in accordance received the method of ovariectomy surgery, but only be resected of small adipose tissue instead of the ovaries.Two and a half months after operation, the rats in aerobic exercise group and aerobic exercise combined the Zuogui Pills started running on the cage, a self-made,40cm in diameter and a counter installed one. The first two weeks were the adaptation period, for the rats learned to run by themselves. Then the exercise training began with 10 minutes running at the 1st day and with 10 minutes increasing in the following days till the running time accumulated to one hour. The number of running circes in the cage were automatically record.3 months postoperation, rats in the aerobic exercise group and aerobic exercise combined Zuogui Pills group took the initiative running for 200 circles in the cage daily, then 3 hours latter, were intragastric administrated by distilled water or Zuogui Pills respectively. The exercise and treatment continued for 12 weeks with six days interruption except one day rest. Rats in model group, estrogen group and Zuogui Pills group also were fed with distilled water, estrogen, and Zuogui Pills separately in the above-mentioned time, which also continued for 12 weeks. Rats in the sham-operation group and the normal control group were fed with equal volume of distilled water in the 12 weeks observed period.Twelve weeks later, rats were anesthetized by intraperitoneal injection of sodium pentobarbital of 45mg/kg body weight, and then quickly decapitated after the blood was taken off from femoral artery. Put the blood standing for 20min, then got the supernatant by being centrifuged (2000 rev/min). Placed the serum obtained by centrifugation in EP tube in the -20℃refrigerator for the CT, BGP, PTH and E2 detection. And the right femurs were collected and prepared for the femoral head shearing test.Results1 Changes of bone biomechanics The maximum load power of the femoral heads of the rats in the model group obviously decreased than that of the normal and sham-operation groups (P<0.01). Compared with that of the model group, the maximum load power of the femoral heads of the rats in the estrogen group, Zuogui Pills group and aerobic exercise combined Zuogui Pills group obviously increased (P<0.01), and the increased extent of the aerobic exercise combined Zuogui Pills group was the highest (P<0.01).2 Hormons examinationCompared with that of normal control group and sham-operation group, osteocalcin (BGP) and parathyroid hormone (PTH) of the rats in the model group elevated. At the same time serum calcitonin (CT) and estradiol (E2) significantly decreased.Compared with that of model group, CT of rats in the zuogui pills group increased, but BGP decreased, CT and E2 increased accompanied with BGP decreased in rats of the aerobic exercise group, BGP and PTH of rats in the Aerobic exercise combined zuogui pills group decreased, but CT and E2 elevated.The comparison among the rats of the aerobic exercise group, Zuogui Pills group and aerobic exercise combined Zuogui Pills group showed that E2 of the rats in aerobic exercise combined Zuogui Pills group significantly increased.Compared with that of model group, BGP and PTH of rats in the estrogen group were significantly lowered, CT and E2 were significantly increased. While compared with that of the other groups, CT, BGP, PTH and E2 of rats in the estrogen group were significantly different.Conclusion1 Aerobic exercise combined Zuogui Pills could improve the bone biomechanics properties, might related with its effect on the balance between bone resorption and formation through the regulating on the key couple signal factors of the bone metabolism and inhibiting bone resorption, to perfect the structure of the bone.2 After taking the Zuogui Pills, taking aerobic exercise, as well as taking both Zuogui Pills and aerobic exercise, serum CT, BGP, PTH and E2 changed in different extents of the rats.3 Taking the Zuogui Pills, taking aerobic exercise, as well as taking both Zuogui Pills and aerobic exercise could make an effect on the osteoporosis treatment of ovariectomized rats. However, there were some differences among the treatment mechanism, and the program of taking both aerobic exercise and Zuogui Pills made a more outstanding effect than the programs of simply taking the Zuogui Pills or simply taking aerobic exercise.
