Previously,we only know the function of alpha-lipoic acid as the effect of energy metabolism and therapy for some illness.more recently,a great deal of attention has been given to antioxidant functions of alpha-lipoic acid by international biologists and medicinists.strenuous exercise increase the producing of free radical and cause disturbance of intracellular prooxidant-antioxidant homeostasis,initiate LPO and destroy the structure and function of tissue and organ, induce exercise fatigue.hence, the supplement of natural antioxidant play an important role on preventing exercise-induced free radical injure and improving the function of antioxidant. Glutathione,which is a important antioxidant,play an important role on maintaining redox state,the experiment is an long-term endurance training exercise model and observe in this exercise model's, effects of Alpha-lipoic acid on Glutathione antioxidant system of skeletal muscel,myocardium,Liver after exhaustive exercise on training Mice and discuss the possible mechanism to the effect of Alpha-lipoic acid on Glutathione antioxidant system in long-term endurance training situation.Putting 32 night-week-old male mice randomly divide into control group(C), exercise group (T), exercise& LA supplymented group (TLA) and LA supplymented group(LA),all the mice engaged in three-day adaptable swimming after raise one week .the mice of C and T group raise quietly, are administered intragastrically to mice daily 0.4ml physiological saline.T and TLA group carry out swimming training for 6 weeks, every week training 6 day.A solution of mixture of LA was prepared in alkaline physiological saline, adjust to pH 7.4 by Hcl, are administered intragastrically to the mice of LA and TLA group daily (100 mg-kg body wt-1day-1) for 6 week before 1 hour in training.the weight of mice is get on Thursday and Sunday.all group which swim on the morning of the day after 6 weeks,are killed after exhaustive exercise to get skeletal muscel, myocardium. Liver to measure the activity of GSII, GPX, GST, GR and MDA.Results: (1)afler long-term exhaustive endurance training, the GSH values in liver, compare TLA group (67.022 ± 7.892 mg/mgprotein) wih T group(59.237 ±5.547 mg/mgprotein), there is significant difference in the statistics results (p=0.041<0.05) ; compare C group (54.675 ± 2.861 mg/mgprotein) wih LA group(63.294 ±6.888 mg/mgprotein), there is also significant difference in the statistics results (p=0.01<0.05) .In skeletal muscel and myocardium,there is no significant difference in the statistics results in every group,but exogenously supplied alpha-lipoic acid decrease exercise-induce GSH loss.compare TLA group and T group, there is no significant difference in the statistics results.(2) after long-term exhaustive endurance training, the GPX values in liver,TLA group value is 56.789 ±3.951 U/mgprotein. compare TLA group with C group (47.604 ± 6.552 U/mgprotein)and T group(51.226 ± 5. 621 U/mgprotein), there is significant difference in the statistics results (P = 0. 001<0.05, P=0.029<0.05) . In skeletal muscel and myocardium.althought there is no significant difference in the statistics results in every group,the activity of GPX in myocardium in TLA group is higher than T group remarkably (P = 0. 078>0. 05) . (3) after long-term exhaustive endurance training, the GST values in myocardium, TLA group value is 44.935 +3.753U/mgprotein. compare TLA group with C group , LA group and T group increase 7.41% , 8.76% and 9.41% respectively, there is significant difference in the statistics results (P = 0. 04X0. 05, P=0. 028<0. 05) . In skeletal muscel and liver, there is no significant difference in the statistics results in every group (P>0.05) It appears that exogenously supplied alpha-lipoic acid decrease exercise-induce the decline of the activity of GST .(4)after long-term exhaustive endurance training ,the GR values in myocardium, compare TLA group...
