The investigation of epidemic disease shows that in recent years, with the improvement of the standard of living and the change of the diet habit, especially the absorption in lots of fat and the reduction of sports, the incidence of cardiovascular disease, for example, the atherosclerosis which is caused by Hypercholesterolemic, has the tendency to increase ,and the serious ones may develop into fatty liver. It has not only high incidence of the. disease among the middle-aged and old-aged, but also has the tendency to increase among the fat children. There are many factors which can cause Hypercholesterolemic, one of the important factors is the abnormal metabolism of Hypercholesterolemic. Liver which is the biggest digestive gland of the human body has the function of secreting bile, processing, compounding and stocking nutritious material, and detoxifying harmful substances from human body and so on. Fatty liver is not a static benign pathological change. If it is delayed and not cured, it may develop into liver fibrosis, cirrhosis of the liver, and even liver function failure, directly threaten the health of the human body, and there is no specific drug in curing fatty liver at present. The contemporary medicine clarifies the mechanism of the disease coming on from the point of pathology physiology to discover the corresponding prophylactic and treatment. The discovery of statins, fenofibrate and the application of therapy contributes a great deal to the prophylactic and treatment of the arteriosclerosis, but there still exist some problems.Isoflavone chemical compound which is one of the active compositions of the natural products has high medical value. Formononetin belongs to isoflavone chemical compound. Formononetin is widely dispersed among the edible plants and medical plants such as peas, cowpeas, soybeans, glycyrrhiza pallidiflora maxim, radix puerar lobato and trifolium pretense, and it has the function of dropping hyperlipklemia, resisting tumor, antilipoperoxidation, anti-osteoporosis, inhibiting helicobacter pylori and so on. But the function which can drop Hypercholesterolemi have not been reported. Because of the low aqueous solution and liposoluable, Formononetin chemical compound as medicine produces low biological utilization ratio and slow results can only be made into the pills. Therefore, the experiments will make Formononetin sulfonated, introduce sulfonic into the molecular structure to raise their aqueous solution, build the animal model of the fatty liver of the experimental rats, observe the function and effect of Formononetin in curingHypercholesterolemic, fatty liver, thus providing theoretical basis for developing the keep-fit food in fatty liver resistance from the influence of Hypercholesterolemic, fatty live, liver function and pathology.Method:70 two-month-old male SD rats(provided by Shaanxi Traditional Chinese Medicine Science Research Institute), fed on standard national rodent dry fodder, having food and drink freely between the temperature of 17 and 23 and the moisture content of 40% and 60%. The experiment will be carried on after one week's adaptable feeding. High fat food is used to induce the rats' high fat model. The experiment is divided into the normal contrast team (A), the model team (B), the normal contrast team plus sports (C), the model team plus Formononetin (D), the model team plus Formononetin plus sports (E), the normal contrast plus Formononetin(F), the model team plussimvastatin(G). There are all 7 teams. The sports loads consult Bedford's research (1979). The training frequency is 5d/week,the increase speed 3m/min, the time 5 mins. The sports increases to the speed of 20m/min, the time is 45-60 mins, then increases the slop to 5%. The whole training process can be stimulated by electricity to force the rats to have the slop training(electric stimulation<1mA). The whole process lasts 8 weeks. At the end of the 8th week all the rats are not fed on in twelve hours and the next day to draw materials, prepare serum, plasma, homogenation part of the liver tissue, observe the influence of Formononetin and sports on TC, TG, HDL-C, AngⅡ, ET, blood T, C, Lep in serum, the content of TC, TG in liver homogenatio; the activation of ALT, AST, ALP, GGT; part of the liver tissue is fixed by formaldehyde then section it to observe the change of liver tissue morphology and the chemical analysis of immunity tissue.Result:1 Hyperlipid-diet increased TC, TG, LDL and droped HDL of the rat blood lipidand, TC, TG, LDL droped amd HDL increased after sports training and Formononetin, at the same time the APOA increased, APOB and lep droped, this imply the lipid metabolism effects of sports trainning and Formononetin is relative to serum polipoprotein and lep.2 Hyperlipid-diet induced TC, TG of liver homogenatio, the liver function dropped(ALT and AST decreased), these all dropped after sports trainning and Formononetin, liver function and is Correspondence to liver lipidand, the effect of Formononetin is obvious than sports trainning. The result implicate the sports trainning and Formononetin can protect and treat liver fatty, improve liver function, the liver function improvement is relative to liver lipidand.3 Hyperlipid-diet induced the serum AngⅡand ET increase, these two dropped after sports trainning and Formononetin, this express that the sports trainning and Formononetin can protect the VEC of the Hyperlipid-diet rat. at the same timethe serum T dropped,this imply that the sports trainning and Formononetin could protect and treat damagement of VEC, which caused by increased T.4 Hyperlipid-diet increased the Bcl-2, Bax, NF-κB and UCP-2 of liver tissues. the liver cells apoptosis dropped, the inflammation factor was suppressed and the liver function protected, because of Bax, NF-κB dropped after the sports trainning and Formononetin, the liver fatty metabolism was adjusted, the high energy was alleviated and the cell inside environment restored dynamic balance due to the UCP-2 decreased. So the reduce fatty and peotect liver function effect of the sports trainning and Formononetin are relative to the AngⅡ, ET Bcl-2, Bax, NF-κB and UCP-2.Conclusion: Formononetin sports can obviously drop the content of TC in the serum and the liver of high fat rats, alleviate the fat drop drops accumlation in the liver cells, and have the function of dropping fat and cultivating the liver, the reduce fatty and peotect liver function effect of the sports trainning and Formononetin are relative to the AngⅡ, ET Bcl-2, Bax, NF-κB and UCP-2...
