Objective:To preliminary study of the changes of mitochondrial function in a state of exercise-induced fatigue and the cause of the changes and to attempts to postpone exercise-induced fatigue based on the principle of the chinese medicine.Methods:A Wistar rat model of intensive swimming training with increasing load by degrees for six weeks was conducted.50 Wistar rats were randomly divided into 5 groups as follows:Sedentary control group(S),Training group(T),Training+astragalus root group(TA), Training+longan with high dosage group(TB),Training+ longan with low dosage group(TC).The rats underwent 6 weeks of swimming training.To reseach the effect of this kind sport mode and chinese traditional medicine to mitochondrial MDA content,SOD and GSH-PX activity,PTP,free calcium content,the velocity of calcium uptaking.Results:(1)After six weeks of swimming training,mitochondrial MDA content of T group was significantly higher than S group(p<0.01),SOD and GSH-PX activity decreased significantly(p<0.05);mitochondrial MDA content of TA and TB group were significantly lower than T group(p<0.01).SOD and GSH-PX activity increased significantly(p<0.05).There was no significant difference between T group and TC group.(2)After six weeks of swimming training,mitochondrial PTP opening of T group significantly increased than S group(p<0.05);TA and TB group mitochondrial PTP opening were significantly lower than T group (p<0.05);There was no significant difference between T group and TC group.(3)After six weeks of swimming training,mitochondrial Ca2+ content and the velocity of mitochondria calcium uptake were significantly lower than T group(p<0.01);mitochondrial Ca2+ content and the velocity of mitochondria calcium uptake of TA group and TB group increased significantly(p<0.05);There was no significant difference between T group and TC group. radicals metabolization in mitochondrial of rats' skeletal muscle.Then these induce largely opening of mitochondria PTP and free calcium trasfer out of mitochondrial.All these leads to the change of mitochondrial structure and function which induce the exercise-induced fatigue. Astragalus root and longan with high dosage could protect mitochondria from being attacted by free radical and have a active effect on stabilizing mitochondrial calcium balance.So Astragalus root and longan with high dosage could protect the integrality of mitochondrial structure and function and have a significant effect on preventing exercise-induced fatigue.Longan with low dosage has no effect on protecting mitochondrial structure and function.
