Background:Brain natriuretic peptide (BNP) is the most popular one in recent years, as a cardiovascular biological marker of clinical application research of BNP for heart failure, the diagnosis, differential diagnosis, treatment, and the risk stratification of monitoring and prognostic. BNP has the value of increasingly reflect. Tumor necrosis factor alpha(TNF-α) is a kind of mononuclear cells and macrophages which secrete a kind of multi-functional of cytokines. TNF-αhas close relationship with the movement. TNF alpha level closely related to the development of CHF. Tumor necrosis factor alpha is one evaluation index of the heart. This experiment used the intraperitoneal injection to obtain simulated chronic heart failure (CHF), and can be used for sports development of CHF intervention study rat model. Through the observation after six weeks swimming training, BNP and TNF contents of the rats would be measured, and further discussed the indicated function in the pathological process with TNF and BNP. This study would provide new insight and important therapeutic bases preventing congestive heart failure. Methods: Seventy-two male SD rats weighing 250-300g were used in this study. Congestive heart failure was produced by the ADR using through intraperitoneal injection. Rats were randomized to untreated infracted group (CHF) or control group (which were sham-operated with same PH indicator and same dose). 5 weeks after, the ECG monitor showed that the CHF group was reacted. Rats were divided into 4 teams: CHF non-movement group, CHF movement group, healthy non-movement group and healthy movement group as the two control groups,18 rats each group. CHF movement group and healthy movement group began to exercise training. After two weeks,these rats were sacrificed. The weight of rats body and heart were measured. All results including heart function, heart blood index and heart muscle for HE. ELISA tests to detect the circulation level of myocardial TNF-αand BNP.Results:①Compared with the control group, the heart weight, cardiac index of infracted group has a very significant increase(P<0.01); LVSP, LVEDP, -dp/dtmax also have a very significant increase(P<0.01)②Heart weight, cardiac index, LVEDP in the CHF non-movement group were significantly lower compared with the CHF movement group(P<0.05), -dp/dtmax in the CHF non-movement group was significantly higher compared with the CHF movement group(P<0.01); compared with the healthy movement group, heart weight, cardiac index is still significantly higher(P<0.05), LVSP is significantly lower than healthy movement group (P<0.01), LVEDP is significantly higher than the movement group(P<0.01)③After six weeks of the exercise, serum BNP of the healthy movement group increased slightly with the movement, but no significant difference(P>0.05); compared with the healthy non-movement group, healthy movement group measured serum BNP increased over the No. 0, 2, 4, 6 weekend, but no significant difference(P>0.05); CHF non-movement group in the six-week exercise of serum BNP increased slightly(P>0.05); No.0, 2, 4, 6 weeks CHF non-movement group measured by the concentration of serum BNP were higher than healthy non-movement group, and there are very significant difference(P<0.01); CHF movement group serum BNP changed little in the six weeks(P>0.05); No. 0, 2, 4, 6 weeks with CHF movement group measured serum BNP levels were significantly higher than the CHF non-movement group(P<0.05)④serum TNF-αof the healthy movement group in the process of exercise in the first 2, 4 weeks increased slightly, the first 6 weeks slightly decreased, but no significant difference(P>0.05); The serum TNF-αbetween the healthy non-movement group and the healthy movement group in the No.0, 2, 4, 6 weekend measured similar(P>0.05); Serum TNF-αin the six-week exercise no change in the CHF non-movement group(P>0.05); The concentration of serum TNF-αof the CHF non-movement group were higher than the healthy non-movement group, and there are very significant difference(P<0.01); Serum TNF-αof the CHF movement group changed little in the six-week exercise. serum TNF-αconcentrations of the CHF movement group were significantly lower than the CHF non-movement group(P<0.05).Conclusion:①The left ventricular diastolic function of CHF rats are significantly decreased after ADR injection, CHF rats modeling success.②Although exercise in rats with heart failure can be significantly improved cardiac index and heart function, but could not to reach the normal range.③BNP and TNF-αwas sensitive when rat ventricular pressure changed, and was significant degree of difference after aerobic exercise.
