Geniposide is an high content active compound in Gardenia fruit, and it has various biological activities in clinical test. Geniposide is an efficient monomer which can be used to produce the gardenia blue pigment and the gardenia red pigment, cross-linking agent natural genipin is the geniposide hydrolysate by the enzyme’s catalytic action.How to separate and purify the geniposide efficiently from the gardenia fruit were reported in this paper. The effect of the isolation of geniposide was investigated with the organic solvent extraction and the macroporous resins. First, geniposide was enriched from the extractum of gardenia fruit with ethyl acetate:n-butyl alcohol(60:40, v/v), the enrichment of geniposide was dissolved in water and then absorbed by D101macroporous resins. The macroporous resins column was eluted with6bed volumes distilled water firstly, then with6bed volumes ethanol-water solution (25%ethanol). The crude geniposide product was yielded by concentrated the eluent of ethanol-water solution (25%ethanol). The98.6%geniposide was obtained after recrystal.The high efficiency of the isolation of geniposide was investigated with the organic solvent extraction and the active carbon in alkaline solution. The acidic substance was transformed into organic salt when the extractum of gardenia fruit’s PH was changed into11. Geniposide was separated from acidic substance efficiently and rapidly when the extractum of gardenia fruit was extracted with n-butyl alcohol, then the n-butyl alcohol extraction was obtained. The n-butyl alcohol extraction was concentrated to copy after decolorized by active carbon. The geniposide precipitate was gained when the ethyl acetate was dropped into the copy substance. The99%geniposide was obtained after the geniposide precipitate was washed two times with ethyl acetate. This technology was best among the document literature. Geniposide was produced vastly and the loss rate was low. The Short Cycle and the lowcost was contained in the technology.
