Gardenia fruit is a dual-purpose plant of herb and food which has been issuedas the first batch resource by Ministry of Public Health.There were variousbiological active compnents in Gardenia fruit.It’s main components were iridoidglycosides(focus on geniposide)and carotenoids(focus on crocins).Gardeniafruit were rich in edible and pharmaceutical value and has broad applicationforeground.The paper researched on the relationship between the main effect ofeach active ingredient of gardenia fruit from different sources and gardeniafood/drug dual characteristics by the chromatography-mass spectrometryfingerprint technology, similarity calculation and Z-score comprehensiveevaluation.It was first to establish differentiated quality evaluation system ofdifferent categories of gardenia fruit raw. Extraction process parameters andextraction solvent of efficient extraction gardenia major functional ingredients werestudied by response surface experiment.The separation and purification techniqueof large-scale preparation of medium-purity geniposide were discussed by two-stepmacroporous resin method. The main results are as follows:(1)It’s used HPLC-DAD method to establish the fingerprints of iridoidglycosides and carotenoids in different producing areas and parts(seeds,peels),by which the similarity evaluation of main components in different producing areaswere calculated. Results show that,species and the main constituent of iridoidglycosides(iridoid glycosides I-V)and carotenoids(carotenoids I-IV)in gardeniafruits were basically the same,high similarities(﹥0.95) are obtained in differentgardenia fruits.The fingerprints of238nm and440nm in different producing areas and partsshow that the species,contents of iridoid glycosides have significant difference inseeds and peels,which was the key factor in pharmaceutical phylogenetic propertyevaluation of Gardenia.Gardenia seeds mainly contains iridoid glycosides III-V.Gardenia peels mainly contains iridoid glycosides I-V.The differentiatedanalysis of fingerprints of iridoid glycosides in different producing areas and partshowed that Gardenia of GuangZhou LuZhai were the same genus,HuBeiChangYang,AnHui BoZhou and ZheJiang AnJie were another same genus,NanYang TangHe was a single genu.(2)The contents of effective component(geniposide,crocins,flavonoids,gardenia oil)of gardenia in different producing areas and parts(seeds,peels)weredetermined.Results show that,Species of iridoid glycosides and carotenoids ingardenia fruits were basically the same.The content of iridoid glycosides andcarotenoids were different,seeds was the highest,peels was the lowest.Gaedenia of HuBei ChangYang and GuangXi LuZhai which were rich ingeniposide as drug intermadeates were the optimal resource.Gaedenia of HuBeiChangYang and Hubei which were rich in crocins as natural pigments were theoptimal resource.Geniposide and crocins content of HuBei ChangYang were thehighest.Flavonoids content of JiangXi FuZhou was more than other producingareas.Gardenia oil content of AnHui BoZhou was highest.Flavonoids content of‘peels’was the highest,and‘seeds’ was the lowest.Gardenia oil content of seedswas the highest.It was researched on the quality of gardenia by measuring the content ofseveral index ingredients.The data were analyzed by adopting Z score multi-indextest and the best gardenia were choosed HuBei ChangYang.Effective componentscontent of HuBei ChangYang were higher than other producing areas.(3)The extraction conditions of crocins and solubilizer were studied by thesingle factors experiment and response surface experiment. The results indicatedthat the optimum extraction conditions were follows: exrtration temperature41.39℃, exrtration time0.75h, Ethanol concentration53.76%. Under the aboveconditions, he extraction yield of croncins was89.64%(the extraction yield of geniposides was89.64%under the same conditions). MES was the best solubilizerto improve the extraction yield of croncins increased by5%.(4)The static adsorption,desorption capacities and of geniposide and crocinson twelve macroporous resins were studied. On the basis,HP-20resin was selectedthe frist resin to separate and purificate geniposide and croncins.Results showthat,60%~80%ethanol was the desorption to eluent crocins.The purity ofcroncins was80%,the color value was380and OD value was less than0.4.20%~25%ethanol was the desorption to eluent geniposide and the purity was33.33%.X-5resin was selected the second resin to purificate geniposide too(pH5.5,20%ethanol)and the purity was65%....
