Gardenia fruit is widely used as a natural food colorant and as a traditional Chinese medicine for treatment of jaundice hepatitis, hypertension and diabetes. This paper mainly dealt with the extraction and purification conditions of gardenia yellow and stability of the colorant. The study laid theoretical foundation for large-scale extraction and purification of gardenia yellow. In addition, the waste solution of the extraction of gardenia yellow is reclaimed to separate effective composition-geniposide, it is very important to utilize synthetically and protect environment.Through experiments, the optimum extraction conditions of ethanol extraction and ultrasonic extraction were obtained. The optimum conditions of ethanol extraction were ethanol/water (6:4), extraction time 2h at 50℃, gardenia fruit :solvent =1:10. As a result, the yield and the color value of the extracted pigment achieved88.81% and 50.19, respectively. The optimum conditions of ultrasonic extraction were ethanol/water (5:5), extraction time 1h at 20℃, gardenia fruit :solvent =1:12. As a result, the yield and the color value of the extracted pigment achieved 98.84% and 70.72, respectively.In this paper, the separation result of the crocin with macroporous adsorption resins column chromatography was studied. Some adsorption resins were chosen for the purpose of separating crocin. By the experiments on the adsorption capacity of static unit volume and the selectivity of every kind of resins for crocin, AB-8 macropore adsorption resin was chosen as the adsorbent of the crocin. Regarding AB-8 macroporous adsorption resin as the research object, the experiments on the static adsorption rate of the resin for crocin, the adsorption curve of the resin for crocin, the flow rate on adsorption , the flow rate on desorption and the solvent of desorption on the behavior of the resin adsorption were made and better conditions of AB-8 absorbing crocin were obtained. While 1.5mL/min as the flow rate onadsorption, 2mL/min as the flow rate on desorption and stepwise elution were adopted, the A238nJA440nm was lowed to 0.22 from 2.90, the recovery was up to 89.27%, and colourity was 221.24, respectively.In addition, the decolor capability of waste solution of gardenia yellow with YWD03K macroporous adsorption resins was discussed. The separation result of geniposide with HPD-400 macroporous adsorption resins by column chromatography was studied. Through the experiments on the static adsorption rate of the HPD-400 resin for geniposide, the adsorption curve of the HPD-400 resin for geniposide and the behavior of the resin adsorption in the flow rate on adsorption, the optimal conditions of HPD-400 resin absorbing geniposide were obtained. After studied the wash of the impurity and the product on adsorption column, the purity of geniposide is more than 80% after drying geniposide solution.
