Aloe is a multifunctional economic crop which has widely applied in medical, cosmetic, food and other areas, and the industrial system has covered cultivation, raw material processing, end product manufacturing and many other aspects. Currently, polysaccharide content of aloe was the primary principle to evaluate the quality of aloe products. Due to the little degree of differentiation between polysaccharide of aloe and polysaccharide of other plants or microbial sources, some merchants often add maltodextrin in aloe products in order to improve the content of polysaccharides in their aloe products, which resulted in a flooding of free aloe products or adulterated aloe products in the market. Therefore, it is urgent to establish a fingerprint identification technology of the characteristic components of aloe vera gel and the techniques and methods to identify the authenticity and quality aloe vera products, and achieve the goal of promoting the healthy development of the aloe industry.With aloe vera gel as the research object, this paper makes a conclusion that the chemical-composition of aloe vera gel can be classified into two categories: macromolecules (polysaccharides) and small molecules (anthraquinone), and then establishes GPC fingerprint and HPLC fingerprint, and combination of these two can be applied to analysis and evaluate the quality of aloe vera gel products.Firstly, this paper studies the GPC analysis conditions of polysaccharides of Aloe vera gel and the identification methods and techniques of GPC standards Fingerprints of aloe vera gel samples from same areas and different areas respectively. Determine the best conditions for GPC measurement were:Gel Permeation Chromatography-Differential Refractive Index Detector analysis system, column:KS-805and KS-803columns, mobile phase:0.1mol/LNaNO3solution (containing0.2%o NaN3), column temperature:60℃, detector temperature:50℃, flow rate:0.8ml/min. Under the certain GPC analysis conditions, this paper establishes the GPC standard fingerprint of aloe vera gel polysaccharides from Beijing and10another regions of China, and analyzes its applicability. The results showed that the similarity of fingerprints of10samples from Beijing region reached up to0.996, similarity of fingerprints of samples from different regions also reached up to0.996. Both were corresponded with the relevant provisions of "technical requirements of fingerprints of Chinese medicine injections (provisional)".Secondly, this paper studies the HPLC analysis conditions of small molecules composition of Aloe vera gel of and the identification methods and techniques of HPLC standards Fingerprints of aloe vera gel samples from same areas and different areas respectively. Determine the best conditions for HPLC measurement were:HPLC-DAD analysis system, column:AgelaVenusil MP-C18column (4.6x250mm,5μm), flow rate of 0.2%acetic acid-methanol:1.0ml/min, mobile phase(amount of methanol):5%(0min)-10%(10min)-40%(20min)-60%(40min), column temperature:35℃, detection wavelength:254nm, injection volume:5μL. Under the defined HPLC analysis conditions, this paper establishes the HPLC standard fingerprint of aloe vera gel small molecular components from Beijing and10another regions of China, and analyzes its applicability. The results showed that the similarity of fingerprints between of10batches of samples from Beijing area were greater than0.977, which is in line with the relevant provisions in " technical requirements of fingerprints of Chinese medicine injections (Provisional)"; similarity of fingerprint between the samples from different regions was greater than0.856, but taking into account of the difference impacts on the accumulation of aloe vera gel composition from weather conditions of different regions such as temperature, humidity, light. By using HPLC fingerprint as the judgment to define quality of aloe products from different regions, the judge criteria can be curtain as the similarity is greater than0.8.
