This paper focused on the extraction of cyclic adenosine 3’,5’-monophos phate (cAMP) and its purification,the optimal conditions are develoved,and extracting of rutin from jujube fruit residue.using ethanol precipitation,ion exchange resin,meme brane, preparative chromatography methods to purify cyclic adenosine 3’,5’-mono-phosphate (cAMP).The aim of comprehensive utilization of jujube is obtaincd.The conclusion of this research were follows:1. establishing method of the high-liquid chromatographic(HPLC) for the analysis of cyclic adenosine 3’,5’-monophosphate(cAMP).Venusil Mp—C18 column (150×4.6 mm,5um) is used. The mobile phase is as follows:V(methanol):V(0.05 M KH2PO4)=20:80,and the flow rate was 1.0ml/min,temperature of column is 35℃,quatity of injection is 20ul.It is measured by an ultraviolet detector under 258.0nm×0.2AUFS.2. Ultrasound combined with the use of microwave extraction method to determine the optimum extraction parameters:ratio of material to liquid 0.07g/ml, microwave power:450W, microwave time of 97s, ultrasonic time of 28min,ultrasonic temperatur-e of 60℃,in these conditions, the test to be gray Xinzheng cyclic adenosine monophosphate date of the extraction 103.15ug/g. In the nine analyzed ZiziPus jujube from different Place, the content of neihuang bianhesuan is the highest for 421.22ug/ g, cAMP content is the lowest in Shandong Ziziphus jujuba for95.57ug/g.3. using ethanol precipitation to remove pectic polysaccharide macromolecular material, the extract concentrated to lOmg/ml, ethanol precipitation with 4 times,6 hours after placing chromatography, centrifugal filtration, the use of phenol sulfuric acid method after precipitation of polysaccharide concentration, the calculation of the removal rate of polysaccharide, the polysaccharide of the removal rate of 70%.4. According to the results of saturated experiment,201×4 ion exchange resin is selected as initial separation of cyclic adenosine monophosphate, derived parameters are:concentration of sample solution 40ug/ml, pH=5.0, the sample rate of V= 1.0ml/min, elution rate of V=1.0 ml/min, elution water first and then eluted with 1N formic acid,8min/formic acid eluate collection tubes, the merger control 5-14 cyclic adenosine monophosphate in the possession of several high by cyclic adenosine monophosphate The HPLC purity:36.25%,83.81% recovery rate. Preparation of the use of waters of cyclic adenosine monophosphate system of separation, After separation of the sample column, HPLC detection of the purity of 90% above, indicating that the preparation C18 column for separation of cyclic AMP.7. Using extracted cyclic adenosine monophosphate jujube fruit residue extract rutin and adequate use of resources. Rutin extraction test conditions to determine their response surface optimization of test parameters to be as follows:70% ethanol concentration, ratio of material to liquid 1:40,extraction time is 9h, extraction temperature of 71.00℃, in these conditions, the date of Rutin in Java extraction rate of 154.66mg/100g.
