Oil-Tea camellia seed meal, which is the remaining of the oil tea camellia seed after extracting oil, is an industrial by-product that has an important application value in the production of camellia oil. Our country has the highest production of Camellia seed in the world, at present, camellia seed meal hasn’t been made good use of, most were directly used as pond cleaning agent and fuel, which resulted in enormous waste of resources and heavy environmental pollution. Studies showed that oil-tea camellia seed meal has obvious bacteriostasis, especially very strong cidal effect on Escherichia coli, Staphylococcus aureus and Aspergillus niger, but at home and abroad, further and systematic study on the antibacterial bioactivitiesit of oil tea camellia seed meal is still poor. This article studied the antibacterial active substances in Camellia Cake extraction, separation and antibacterial activity and stability by the antibacterial activity tests.The conclusions are as follows:(1) Using different concentrations of ethanol as extraction medium, we studied the effects of extracting temperature, time, the ratio of material to solvent on extracting the antibacterial bioactivities from tea seed meal.Adopting L9(34) orthogonal test to optimize the extraction technology of antibacterial substance, based on single factor design, we get the optimal extraction process which is ethanol concentration 80%, extraction temperature is 80℃, iquid-solid ratio 6:1, extraction time was 4 h.(2)Using petroleum ether, ethyl acetate and n-butanol to continuously extract the tea seed extracts and mensurate the antibacterial activity of extracts, it found that n-butanol extract has stronger antimicrobial activities than other components,Which has the best antibacterial effect on escherichia coli, Staphylococcus aureus and Aspergillus niger and the results showed that n-butanol was an effective substance used into further purification of crude extract.(3) The contents of the main substances in the crude extraction of Camellia meal were measured, the result is crude fat 3.76% + 0.33, tea polysaccharide 21% + 0.25, flavonoids 5.6% + 0.28, saponin 18.78% + 0.35.(4)N-butyl alcohol extract of oil tea camellia seed meal has broad-spectrum antibacterial activity. It has good inhibitory effect on Escherichia coli, Staphylococcus aureus, Salmonella, Bacillus thuringiensis, Aspergillus, Rhizopus nigricans. The measured minimum inhibitoryconcentrations were escherichia coli 77.815 g/ml, Staphylococcus aureus 83.90691.25 g/ml, Salmonella bacillus thuringiensis 91.25 g/ml, 112.905 g/m,Bacillus thuringiensis 112.905 mu g/ml,Aspergillus niger 267.4 mu g/ml,Rhizopus nigricans 180.3 g/ml using the resazurin assay.And the inhibiting effect on bacteria is better than on molds.(5)Through studying the stability of antibacterial activity of N-butyl alcohol extract of the oil tea camellia seed meal,It showed that in different temperature, illumination, p H value, ultraviolet radiation, the effects of the metal ions, the stability of antibacterial effect is high. For thermal stability, it can tolerate high temperature for a short of time. For p H stability, it is relative more stable under p H6, and its stability under acidic condition is higher than that under alkaline condition. For ionic stability, it presents stable antimicrobial stability in solution of Na+, K+, Ba2+, Mg2+, Ca2+ and Fe3+ respectively.
