In this study,the extraction process,chemical components,fingerprints and Antibacterial activities as well as antioxidation of volatile oil from Rosa multiflora Thunb were investigated.The main contents and conclusions are as follows:(1) GC-MS was used to analyze the chemical constituents of volatile oil from Rosa multiflora Thunb with diffrent distill methods. Results the extraction rates of volatile oil with Soxhlet extraction(SE)was 3.17 %,the components of the volatile oil were examind by GC-MS,73 of the 85 were identified.The extraction rates of volatile oil with ultrasonic wave was 2.08 %,the components examind 39 of the 64 were identified,the extraction rates of volatile oil with Microwave-assisted extraction was3.03%,the components examind 50 of the 71 were identified,Ultrasonic-Micro wave synergistic extraction was 3.43 %,the components examind 69 of the 142 were identified.Conclution the components of volatile oil from Rosa multiflora Thunb with diffrent distill methods have difference but similarities,it can provide a method for Rosa multiflora Thunbs quality control,the extraction rates is higher and the components are more with the method of ultrasonic-microwave synergistic Extraction.(2) To study the Optimization of ultrasonic-microwave synergistic Extraction Technology for Volatile Oil from Rosa multiflora Thunband gas chromato Graphy mass spectrometry(GC-MS)Analysis of Its chemical composition.Methods:The influences of solvent to material ratio,microwave power and extraction time on the extraction yield of volatile oil from Rosa multiflora Thunb with the ultrasonicmicrowave synergistic extraction(UMSE)technique were studied.Box-Behnken experimental design was based on the single factor experiments.One mathematical model established and analyzed by response surface methodology(RSM)was adequate to describe the relationships between the studied factors and the response of volatile oil extraction yield.Based on the canonical analysis,the optimum extraction conditions were obtained as follows:solvent to material ratio 9.88 m L/g,microwave power 276 W and extraction time156 s.Under the optimized extraction conditions,the extraction yield is 3.82 %,which is close to the estimated value 3.88 % attained by using regression model.This model can well predict the extraction ratio of Volatile Oil from Rosa multiflora Thunb Then the chemical components of the volatile oil were examined by GC-MS. Finally 104 kinds of compounds were separated,49 kinds of compounds were identified,and the relative contents of these components were determined by peak area normalization method.(3) GC was adopted to build the standard fingerprints of volatile oil from rosa multiflora thunb for the 12 habitats,which identified 14 components,as the indicator component of the volatiel oil from rose multiflora to identify the standard,The GC analysis condition of the fingerprints has the advantage of very good separating effect,the similarity of the 12 volatile oil from rose multiflora thunb is higher than0.84,the precision,stability and repeatablity of the fingerprints were better that the fingerprints can be used as the standard and the quality control method of the medical materials from rose multiflora thunb.(4) Filter paper measuration was adopted to estimate the antibacterial activity of the volatile oil from rose multiflora thumb,the results showed that volatile oil samples from rose multiflora thunb have certain degree of growth inhibition to staphylococcus aureus,bacillus subtilis,escherichia coli,degeneration bacillus,white slime mold,their antibacterial mechanism may be related to chemical components contained in the volatile oil,such as guaiol,humulene,carvone,caryophylleneeu,genol.(5) Take volatile oil from rose multiflora thunb and Vc as control group,use spectrophotometric method to determine the clearance rates of DPPH·and·OH,as well as superoxide anion,the results show that the effects of volatile oil of rose multiflora thunb on the clearance rate or inhibition ratio of DPPH· and·OH,as well as superoxide anion are better than control group and the reducibility of them is weaker than Vc,but which overgrows following with the concentration of volatile oil.
