Studies on Related Techniques of Tissue Culture aiid Rapid Propagation of Phalaenophsis hybrid and Gldiolus hybridus ABSTRACT Selected suitable explants from flower peduncle terminal buds and flower peduncle axillary buds of Phalaenophsis hybrid and young dormant corms and new corrn buds of Gldiolus hybridus were cultured on different culture media, and various factors affecting tissue culture were also studied systematically. Results of tissue culture of P hybrid showed that (1) axillarv shoot were better than terminal buds and (2) the induced rate of lateral buds in central position of flower peduncle were higher than the others, especially the second flower peduncle axillary shoot had the highest induced rate; However, the top axillary shoot had the lowest induced rate; (3) Pollution rate in explants inoculation was related to the ageing degree of young explants and pollution rate of new flower peduncle was the lowest; (4) Axillary shoots were induced well. on MS medium supplied BA 3.OmglL and NAA l.Omg/L for primary culture and with BA 5.Omg/L and NAA 1 .Omg/L for subculture; Besides, MS medium containing activated carbon O.6gfL was suitable rooting-medium. It has been concluded from tissue culture of G. hybridus that (5) young dormant cms and new corm buds of G. hybridus were cultured better than cut sections of corms (no buds); (6) Rapid propagation clones were gained on modified MS medium; Moreover, after peeling off young dormant corms in primary culture, reproducing rate was the highest on MS medium supplied BA 1 .Orng/L and NAA 1 .Omg/L; Besides, induced rate of callus was the higher on MS medium supplied BA 2.OrngIL and NAA 1 .0mg~t, and the highest induced rate was about 86.0%; (7) The suitable primaly culture media were MS medium supplied BA 3.Orng/L in tissue culture of new corrns buds and (8) MS medium supplied BA 1 .Omg/L were best media for inducing rosette buds and MS medium supplied NAA O.5mgIL were suitable rooting-media.
