Radix puerariae is a traditional Chinese medicine in common use.It is maily the root of Pueraria lobata (Willd.) Ohwi and Pueraria thomsonii Benth. of the genus Pueraria DC.,Pea family and has isoflavones ingredients, such as daidzein,daidzin and puearin, and plenty of starch.In the recent years .people attach much importance to Radix puerariae because the isoflavones ingredients has notable treatmental effect of cardiovascular and cerebrovascular disease and many products made of radix puerariae are presented in the supermarket and hospital and there has a increasing need of it. At present, Pueraria lobata (Willd) Ohwi and Pueraria thomsonii Benth.are the maily species used for medical purpose.But the resource of Pueraria lobata (Willd.) Ohwi is very limited and it would do serious destroy to the ecological environment by digging a lot.On the other hand .the content of total isoflavones of Pueraria thomsonii Benth. is lower than Pueraria lobata (Willd.) Ohwi.So the breeding of autopolyploid would be significant to improve the produce and quality of Pueraria thomsonii Benth..Tissue culture and colchicines induction were applied to the induction of autotetraploid of two large-area-cultivated matierials,Jiangxigeboshi and Hechuangegen, in the study.This would lay foundation for the future selection of new materials with increased produce and top quality.Stem tip .stem and leaf were the explants in tissue culture.The germ-free propagation system was established by regenerating callus or inducing growth of axillary bud.Comparing the effect of different kinds of plant growth regulators and different hormon concentration.the media suitable for the two materials were singled out.Based on the establishment of propagation system,the best fit of optimum concentration and time interval of colchicines processing was screened to process the apexes of plantlets and little plants.The result suggests that: stem tip and stem are better than leaf as explants.both regenerating callus and inducing growth of axillary bud can get propagation system.but the latter can save much time.In the condition of pH 5.6-5.8,concentration of saccharose 3%,temprature 25±2℃ .intensity of illumination 1500-20001x and illumination time 10-12h/d,the best media fitting both materials are MS+1.0mg/L2,4-D+1.0mg/L6-BA+0.2 mg/L NAA,MS+0.2mg/L2,4-D+1.0mg/L6-BA and 1/2MS+0.2 mg/LIBA for callus induction.propagation and rootage repeatedly .The study also reveals that both materials are with great tolerance of colchicines, concentration 0.6% processing 36h and 0.2% processing 72h did not deaden the apexes,and planlet's apex has much higher induction rate than the lymph's apex . So it is better to chosing higher concentration.But the low livability and tardy growth of apex disadvantage the future selection after high-concentration- colchicines treatment.Concentration 0.4%~0.5% processing 48h is the best fit for best induction effect.The highest indution rates of planlet's apex were 29.6% for Jiangxigeboshi and 35.0% for...
