| The studies on improvement, identification and alternative raw materials of good strain of Auricularia auricular (black fungus)are practically significant for guiding production and protecting forest resources. The domestic strain used and alternative culture materials were studied, combining the resources and climate characteristics of Dong-ning county of Heilongjiang Province the major producing areas of Auricularia auricular in China. The 8 wild strains were collected and separated from forests in huma county, jiayin county, fuyuan county and sanchakou town of dongning county of sino-russian frontier. Then the 8 wild strains were test cultured. The growth of mycelium at each level of medium, natural contamination resistance, yield and characteristics of strains were tested. The JY04 from forests of Jiayin County of Heilongjiang province was determined as an excellent strain due to its better performance in each test. The strain was identified in morphology and molecular biology by colony morphology, characteristics of mycelium,18S rDNA gene sequence analysis and so on. The results of 18S rDNA gene sequence analysis showed that the similarity of the 18S rDNA gene of JY04 and Auricularia auricula-judae strain HF-9 18S ribosomal RNA gene (GQ168493.1) in database was 99%.In the alternative raw materials tests, the corncob, beanstalk, mushroom bran and leaf powder of broadleaf, easily available wastes with wide resources, were tested to replace sawdust of broadleaf in normal formula. The contamination, yield and mycelia growth of different formula in two and three grade production and culture tests were studied. The results showed that alternative raw materials added in appropriate proportion was good for mycelium growth promptly rather than be bad for the yield. At last, the best proportion of alternative material was determined as 40% of corncob,40% mushroom bran,50% of beanstalk and 30% of power of broadleaves. The separation, selection, test cultured of study of Auricularia auricular strains make a foundation for improving the yields and quality of Auricularia auricular; it is hope that the problem of increasingly shortage of raw materials can be relieved in a certain extent in the cultivation procedure of Auricularia auricular by the study of alternative raw material, it provided a valuable experience for follow-up studies in alternative raw material and indirectly protect the environment, promoting the heathy development of Auricularia auricular industry in the locality, at the same time, which make a positive contribution for local economic. The completion of this project will have significant economic, social and ecological benefits. |
